摘　　要：[目的]建立一种NK细胞体外选择性高效扩增的实验体系,观察其体内的抗肿瘤活性。[方法]以干细胞培养基(SCGM)为培养基,在低剂量IL-2(100U/ml)联合14mer肽TKD(TKDNNLLGRFELSG,aa450-463)(2μg/ml)的培养条件下,从人外周血PBMC中高效扩增获得CD3-CD56+NK细胞,并建立肝癌裸鼠移植瘤模型,观察NK细胞在体内的抗肿瘤活性。[结果]建立了一种高效的从人外周血PBMC中选择性扩增NK细胞的体系,该NK细胞对表面表达HSP70的肝癌移植瘤模型具有显著的抗肿瘤作用。[结论]经过14mer肽TKD激活的NK细胞对表面表达HSP70的肿瘤具有很强的溶解活性。[Objective] To establish the selective proliferation system in vitro and evaluation of anti-tumor activity in viro of natural killer（NK）cells.[Methods] Human peripheral blood cells（PBMCs）were incubated with stem cell growth medi-um（SCGM）and stimulated by low dosage of Interleukin 2（IL2,100 U /ml）combined TDK oligo peptide（14 mer,2 μg /ml）to develop the NK cells with CD3-CD 56＋phenotype.The naked and hepatocellular carcinoma-bearing mice were used to eval-uate the anti-tumor activity of the NK cells.[Results] The high efficient selective proliferation system of NK cells was estab-lished in vitro by stimulation with TDK oligo peptide,and the cells in mouse model showed that enhanced cytolytic activity against hepatocellular carcinoma cells expressed HSP70 on the membrane.[Conclusion] The NK cells stimulated by the TDK oligo peptide could develop from PBMCs and be enhanced the cytolytic activity against hepatocellular carcinoma cells with HSP70 expressed on the membrane.
