摘 要:目的探讨肺癌组织中缺氧诱导因子-1α(HIF-1α)、葡萄糖转运蛋白1(Glut1)、Glypican-3的表达及其临床意义。方法采用免疫组织化学方法检测52例(男35例,女17例;鳞癌32例,腺癌20例)肺癌组织中HIF-1α、Glut1、Glypican-3的表达。结果 52例肺癌组织中HIF-1α阳性表达36例(69.2%);Glut1阳性表达38例(73.1%);Glypican-3阳性表达7例(13.5%)。肺癌组织Ⅲ~Ⅳ期与Ⅰ~Ⅱ期中高度分化与低分化组比较,HIF-1α、Glut1阳性表达率均明显增加,差异有统计学意义(P<0.05)。肺癌组织有淋巴结转移组与无淋巴结转移组比较,HIF-1α、Glut1阳性表达率均明显增加,差异有统计学意义(P<0.05)。肺癌组织中不同年龄组及不同性别组比较,HIF-1α、Glut1阳性表达差异无统计学意义(P>0.05)。Glypican-3在肺癌组织中与在癌旁组织中阳性表达率差异无统计学意义(P>0.05)。结论肺癌组织中HIF-1α与Glut1蛋白阳性表达,HIF-1α与Glut1阳性表达与肺癌的病理分期、分化程度和淋巴结转移有关,与年龄、性别等无关。Glypican-3在肺癌中表达较低,与癌旁组织中的表达无差异。Objective To explore the expression and clinical significance of hypoxia induced factor 1α(HIF-1α) and glucose transporter 1 (Glut 1) and Glypican-3 in lung carcinoma.Methods The expression of HIF-1αand Glut1 and Glypican-3 were tested by immunohistochemistry in 52 cases of lung cancer specimens (35 males and 17 females;32 cases with squamous carcinoma,20 cases with adenocarcinoma).Results In 52 cases of lung carcinoma specimens,36 cases showed positive staining of HIF-1α protein (69.2% );38 cases showed positive staining of Glut 1 protein (73.1% ).7 cases showed positive staining of Glypican-3 (13.5% ).The expression rate of HIF-1a and Glut1 in lung carcinoma tissues of Ⅲ-Ⅳ stage poorly differentiated groups was significantly higher than that inⅠ-Ⅱstage middle-highly differentiated groups (P0.05 ).The expression of HIF-1a and Glut 1with lymphatic metastasis in lung carcinoma was significantly higher than those without lymphaticmetastasis (P0.05).The expression rate of HIF-1a and Glut1 in lung carcinoma tissues was not significantly higher in different age groups or different sex groups(P0.05).The positive expression of Glypican-3 in lung cancer tissues and non-neoplasia lung tissues was low and the difference between the two groups was not statistically singificant(P0.05).Conclusion High expression of HIF-1αand Glut 1at protein level could be seen in human lung cancer,high expression of HIF-1α was associated with invasion degree and differentiation degree and lymphatic metastasis of lung carcinoma but not with the age or gender of the patients.Low expression of Glypican-3 could be seen in human lung cancer,the difference of expression in lung cancer and in non-neoclassic lung tissues was not significant.
