血卟啉衍生物对γ线照射Eca109细胞的增敏效应

摘  要:人食管癌上皮细胞体外经血卟啉(15μg/ml)与γ线(5Gy)处理后,癌细胞生长受抑,分裂指数明显降低,倍增时间显著延长,形态学观察,细胞重度空泡变性、胞浆凝固、大片坏死,超微结构示线粒体空化、退变,粗面内质网扩张,核糖体脱失,细胞膜破损。FCM分析细胞被阻滞在G_2M期,并杀伤细胞于此期。结果提示血卟啉衍生物具有放射增敏作用,在分子水平上表明了细胞生长受抑的机制。Cultured human esophageal careinoma cells Eca109, were first treated with HPD(15 μg/ml.) and then 5 GyΥ-ray irradiation.The morphological, functional and metabolic changes of the cells were studied by means of light, electron microscope and FCM observation. The cells treated with HPD plus Υ-ray showed that: 1) Cell count,mitotic index were decreased and cell population doubling time was prolonged. 2) Under light microscope,the cell was shown to be first severe vascuolar degeneration and cell naked nuclei and lysis could be often seen. 3) SEM and TEM observations revealed that microvilli on the cell surface was reduced in number and membrane was seen broken. In the cytoplasm,mitochondrias showed degenerating changes.Such as swollen,marrow like body,etc. RER was dilated and ribosomes were lost. 4) FCM analyses indicated the cells were inhibited in G_2M phased. At the same time,cellular fragment increased. All the result suggested that HPD did sensitization enhancement effects for Υ-ray in Eca109 cells.
