摘  要:目的水通道蛋白作为细胞膜特异性水转运蛋白在脑内分布广泛,参与脑内水平衡调节,与脑肿瘤、脑创伤等多种疾病导致的脑水肿有关。本实验主要研究水通道蛋白1、4在培养大鼠星形胶质细胞、人多形性胶质母细胞瘤系BT325细胞及人脑胶质瘤组织中的表达情况。方法应用胶质纤维酸性蛋白(GFAP)免疫细胞化学染色鉴定培养大鼠星形胶质细胞及BT325细胞。应用反转录PCR法检测GFAP、AQP1及AQP4在不同组织细胞中的表达。结果实验中培养大鼠星形胶质细胞及BT325细胞GFAP免疫细胞化学染色阳性。体外培养大鼠星形胶质细胞、人胶质母细胞瘤组织及对照组标本均有GFAP表达,同时也有AQP1及AQP4表达。BT325细胞只有GFAP表达,无AQP1及AQP4表达。结论 AQP1、4在培养大鼠星形胶质细胞及胶质瘤组织中均有表达,可能对脑功能的维持及肿瘤性脑水肿的发生、消散有重要影响。Objective Aquaporins constitute the principal pathway as water moves through cell membrane in brain.They may play central roles in the homeostasis of water content in the brain and brain edema caused by brain tumor or brain trauma.In this study,we investigated the expression of AQP1 and AQP4 mRNA in cultured rat astrocytes,human glioma BT325 cell lines and human glioma.Methods Astrocytes and BT325 were identified by immunostaining with anti-glial fibrillary acidic protein(GFAP) antibody.The expression of GFAP,AQP1 and AQP4 mRNA were detected by reverse transcription polymerase chain reaction in cultured rat astrocytes,BT325 and human glioma.Results The immunostain for GFAP was positive in cultured rat astrocytes and BT325.The expression of GFAP mRNA was detected in cultured rat astrocytes,BT325 and human glioma.The expression of GFAP as well as AQP1 and AQP4 mRNA were found in cultured rat astrocytes,human glioma and the control group,but in BT325,only the expression of GFAP was found.Conclusion The AQP1 and AQP4 are expressed in cultured rat astrocytes and human glioma,they may play important roles in the maintenance of brain function as well as in the occurrence and disappearance of brain tumor edema.
