摘  要:应用离子注入技术,对能转化左旋乙基甾烯双酮为15α-羟基左旋乙基甾烯双酮的雷斯青霉(Penicillium raistrickii)进行研究,旨在提高其C15α位羟化能力。结果表明,雷斯青霉存活率曲线呈典型的"马鞍型",通过液相色谱分析,在N+注入剂量为50×1014ions.cm-2时可引起较高的正突变率,并从中获得一突变株,其15α-羟基左旋乙基甾烯双酮转化率达到62.7%,为出发菌株的1.16倍。In this paper,Penicillium raistrickii,which can transform 13-ethyl-gon-4-en-3,17-dione to 15α-hydroxyl-13-ethyl-gon-4-en-3,l7-dione,was induced by the technology of ion implantation,in order to improve its 15α-hydroxylation ability.The results indicated that the survival rate curve showed typical saddle′s shape.Analyzed by HPLC,there was high positive mutation rate when the N^+ implantation dose was 50×10^14 ions·cm^-2 under implantation energy 30 keV.A mutant was screened out.Its biotransformation rate for the 15α-hydroxylation of 13-ethyl-gon-4-en-3,l7-dione reached 62.7%,which was 1.16 times as much as that of the original strain.
