摘 要:[目的]为了明确合肥地区大麦黄矮病毒株系种类及分子变异情况。[方法]利用生物学手段和RT-PCR方法鉴定了2009年采自合肥地区的19个分离物,并测定了分离物的外壳蛋白基因(CP基因)的序列。[结果]合肥地区的12个分离物被鉴定为BYDV-PAV,CP基因序列比对发现核苷酸一致性为95.8%~99.7%。系统进化树分析得知,12个合肥分离物序列聚类到一个分支上,它们与中国分离物PAV-CN(AY855920)的CP基因亲缘关系最近。[结论]掌握合肥地区大麦黄矮病毒的株系分布及分子变异情况,对指导该地区小麦黄矮病的抗性育种和防治工作有着非常重要的意义。[Objective] The species and molecular variability of the strain of barely yellow dwarf virus(BYDV) isolated in Hefei was clarified. [Method] 19 isolates collected in 2009 in Hefei were identified with the method of biology and RT-PCR,and the sequence of the coat protein gene of the isolates was tested. [Results] 12 isolates were identified as BYDV-PAV and the unanimity of the nucleotide was 95.8%-99.7% compared with the CP gene sequence. It was revealed that 12 isolates from Hefei was clustered into a branch through the analysis of the hylogenetic tree,which was closed to the PAV-CN gene(AF216863) of the isolate of China in genetic relationship. [Conclusion] The distribution of BYDV strain in Hefei and its molecular variability were mastered,which was significant in the guidance to the resistant breeding and the control of the disease.
