摘 要:目的探索多发性骨髓瘤(MM)肿瘤克隆性前体细胞作为MM治疗学上的新的靶细胞的可能性。方法以全反式维甲酸(ATRA)和体外培养的肿瘤克隆性前体细胞为模型,联合应用台盼蓝拒染试验、瘤细胞集落形成试验、光镜下形态学观察、免疫细胞化学染色、扫描电镜技术等方法研究与分析ATRA对肿瘤克隆性前体细胞增殖动力学的影响。结果13例MM患者PBMNCs样本在体外与ATRA作用14天后,有6例样本中显示出ATRA对肿瘤克隆性前体细胞有不同程度的抑制作用,有效率为46.2%。经过ATRA处理过的细胞样本,未见明显的细胞毒作用,培养物中以粒-单细胞集落生长为主,而相应对照组则以瘤细胞集落生长为主。结论ATRA在体外培养系中能明显抑制MM肿瘤克隆性前体细胞的增殖。肿瘤克隆性前体细胞可能是MM治疗学上新的靶细胞。objective To investigate the possibility of novel therapeutic strategy in which tumor clonogenic precursors would be used as target cells for drugs. Methods In vitro cultured tumor clonogenic precursors with ATRA were served as models,and effect of ATRA on the growth of tumor precursors was evaluated by the trypan blue exclusion method,myeloma cell colony formation assay,morphology,immunochemical staining and scanning electron mircroscopy. Results Of 13 samples exposed to ATRA for 14 days,6 cultures showed significant antiproliferative effects of ATRA on tumor precursors,and the response rate was 46.2%.Moreover,no obvious cytotoxicity was found in ATRA treated cultures.GMCFUs were the major cell colonies in ATRA treated cultures,while myeloma cell colonies in corresponding controls.HZConclusion ATRA could inhibited tumor clonogenic precursors of multiple myeloma patients and tumor clonogenic precursors might be used as the target cells in multiple myeloma therapy.
