摘 要:[目的]探究同工酶技术在杏鲍菇和鸡腿菇分类鉴别中的应用效果。[方法]利用垂直板聚丙烯酰胺凝胶电泳,对来源不同的杏鲍菇和鸡腿菇进行酯酶同工酶比较和分析,并通过聚类分析对各菌株间的遗传差异性进行研究。[结果]杏鲍菇电泳共检出18条迁移率不同谱带,聚类分析可分为3类:Pa1与Pa5酶谱相同归为1类;Pa2与Pa3酶谱相同归为1类;Pa4自成1类。鸡腿菇电泳共检出11条谱带,聚类分析可分为3类:Cc1与Cc3酶谱相同归为1类;Cc2与Cc4酶谱相同归为1类;Cc5另成1类。[结论]酯酶同工酶可作为杏鲍菇和鸡腿菇种质资源研究和管理方面的一种有效方法。[Objective]The effect of esterase isoezyme on the classification and identification of Pleurotus eryngii and Coprinus comatus Strains was studied.[Method]With the vertical flat electrophoresis of polyacrylamide gel,Pleurotus eryngii and Coprinus comatus strains from different regions were compared and analyzed in esterase isozyme character and the genetic differences among strainswere analyzed with the cluster analysis software.[Results]There were 18 bands detected with electrophoresis staining,the majority of which showed the difference in isozyme patterns.Pe1 and Pe5 could be separated into first group;Pe2 and Pe3 could be separated into second group;Pe4 could be separated into third group by itself.There were 11 bands detected with electrophoresis staining,the majority of which showed the difference in isozyme patterns.Cc1 and Cc3 could be separated into first group;Cc2 and Cc4 could be separated into second group;Cc5 could be separated into third group by itself.[Conclusion]The esterase isozymes could be used as one of the effective biochemical indicators in the study on the Pleurotus eryngii and Coprinus comatus germplasm resources.
