摘 要:目的用动脉内置线阻断法建立大鼠大脑中动脉(MCA)局灶性脑缺血模型,观察缺血脑区中TNF-α的变化。方法改良MCA缺血模型,设30min缺血后不同时点(6h,12h,24h,48h)再灌注组,采用TNF-α酶联免疫吸附法检测各缺血脑区的TNF水平。结果缺血30min后各时相点中,6h再灌流组缺血侧纹状体和皮层TNF-α水平即明显增加,后略下降,至再灌流48h后出现第2个高峰。结论30min缺血后各时相点的TNF-α出现观相性增高现象。Objective to examine the relase of TNF - a in ischemic cerebrum using reversible focal middle cerebral artery (MCA)ischemia in rat induced by intraluminal vascular occlusion method .Methods 6, 12,24,48 hours reperfusion following 30 -min occlusion using improved focal cerebral ischemia model ,the levels of TNF - a in ischemic cerebrum were detected with Tumor Necrosis Factor-a ELISA Kit.Results The levels of TNF - a in ischemic striatum and cortex obvious increased at 6 hours after 30 - min ischemia and then and then declined . However,it had a second peak following 48 hours reperfusion .Conchusion Increase of the TNF - a displayed biphasic changes in different reperfusion time after 30 - min ischemia .
