摘  要:目的:探讨热休克大鼠小脑小脑肽1(Cbln1)和孤独谷氨酸受体-2(Glud2)表达的变化及意义。方法:60只SD大鼠随机分为3组:空白组,休克早期组和休克晚期组。大鼠置于恒温恒湿箱模拟湿热天气。生理记录仪检测大鼠生命体征。ELISA检测小脑组织中Cbln1和血清中降钙素基因相关肽(CGRP)含量,流式细胞仪检测Glud2表达,放射免疫分析法检测血清中肾上腺素(E)、去甲肾上腺素(NE)、血管紧张素-Ⅱ(AngⅡ)、肾素(RA)浓度。结果:休克早期组和休克晚期组Cbln1和Glud2含量显著高于空白组(P<0.05);休克晚期组Cbln1和Glud2含量显著高于休克早期组(P<0.05);相关性分析显示Cbln1、Glud2分别与E、NE、AngⅡ、RA呈正相关(P<0.05);Cbln1和Glud2与CGRP无明显相关性(P>0.05)。结论:热休克大鼠Cbln1和Glud2表达增加,它们可能是自身抗休克机制之一。Objective:To investigate the expressing of Cerebellin-1(Cbln1)and orphan glutamate receptor-2(Glud 2) in the heat shock rat cerebellum.Methods:Sixty SD rats were randomly divided into control group,early shock group and late shock group.Rats were placed in constant temperature and humidity boxes to simulate the weather of hot and humid.Electrophysiolograph detect the vital sign of the rats.Radioimmunoassay was used to detect the serum level of adrenaline (E),norepinephrine(NE),angiotensin-Ⅱ (AngⅡ),and renin(RA),ELISA was used to determinate the level of Cbln1in the cerebellum and the serum level of calcitonin gene-related peptide (CGRP),Glud2 expression was detected by flow cytometry.Results:Expression of Cbln1and Glud2 in the early shock group and late shock group were significantly higher than that in the control group respectively(all P0.05).Expression of Cbln1 and Glud2 in the late shock group were significantly higher than that in the early shock group(both P0.05).Correlation analysis in Cbln1and Glud2were positively correlated with E,NE,AngⅡ,and RA (P0.05),but no correlation with CGRP(P0.05).Conclusion:The over expression of Cbln1and Glud2in heat shock rats may be one of its mechanism of anti-shock.
