摘  要:目的探讨radixin和moesin是否也和ezrin一样在肿瘤转移中起调控作用。方法选取ERM均表达的人胃癌SGC细胞作为模型,运用RNA干扰(RNAi)方法分别构建ezrin,radixin和moeisn表达特异性下调的稳定细胞株,并比较它们的体外转移能力。结果体外细胞侵袭实验表明ERM中任一蛋白的特异性下调均能减弱细胞的转移能力,Western blot分析显示ERM中的任一蛋白的特异性下调均引起E-cadherin表达的明显增加。结论 3种ERM蛋白在SGC细胞的转移过程中都起重要的作用,下调ERM蛋白中的任一成员都能明显降低SGC细胞的转移能力,ERM与E-cadherin间的相互作用可能在肿瘤的发展过程中至关重要。我们的发现同时也为ERM功能冗余论提供了佐证。Objective To explore whether ERM proteins share the same function in tumor metastasis. Methods In the present study,we chose human gastric carcinoma SGC cell that expressing all ERM proteins as the model to examine their unique roles in this matter.By using the RNA interference(RNAi) method,ezrin,radixin and moesin stable knockdown SGC cells were constructed,which ensured the metastasis assay operated in optimal condition. Results In vitro cell invasion experiments showed that all ERM specific deficiency caused the reduction of metastatic ability.Western blot analysis showed that the expression of E-cadherin significantly increased when any ERM protein was down-regulated. Conclusions Our results indicated that all ERM proteins play similar roles in the SGC cell metastasis,all ERM proteins can reduce the metastatic ability of SGC cell,and the mechanism between ERM and E-cadherin might play an important role in tumor progression.This is a new evidence for ERM functional redundancy.
