摘  要:目的研究负载表柔比星的磁性CS-PAA-EPI纳米粒在人膀胱癌移植肿瘤模型中的抗肿瘤效应。方法选用壳聚糖、磁流体及表柔比星为原材料首次合成负载表柔比星的磁性壳聚糖纳米粒,建立人膀胱癌移植肿瘤模型,通过瘤重抑瘤率、瘤体积抑瘤率及TUNEL等技术检测CS-PAA-EPI磁性纳米粒在人膀胱癌移植肿瘤模型中的抗肿瘤效应。结果在外加磁场的作用下,中剂量CS-PAA-EPI磁性纳米粒组的瘤重抑瘤率、瘤体积抑瘤率及TUNEL凋亡率分别为83.3%、77.8%和75.3%,均明显高于EPI生理盐水组(P<0.01)。结论在外加磁场的作用下,磁性CS-PAA纳米粒载药系统负载表柔比星对人膀胱癌裸鼠移植肿瘤进行局部化疗,其疗效明显优于常规化疗,为膀胱癌治疗提供了新方法。Objective To investigate the effects of CS-PAA-EPI polymer magnetic nanoparticle on human bladder cancer transplanted tumor model. Methods Magnetic CS-PAA-EPI nanopaticle was composed by CS,acrylic monomer(AA),EPI and Fe3O4 nanopaticle.Human bladder cancer transplanted tumor model was established.The tumor weight inhibition rate,the tumor volume inhibition rate and apotosis rate were detected,which was used to evaluate the antitumor effects of CS-PAA-EPI polymer magnetic nanoparticle. Results Compared with control group,the tumor weight inhibition rate(83.3%),the tumor volumet inhibition rate(77.8%) and apotosis rate(75.3%) in experimental group were significantly increased(P0.01). Conclusions The effect of CS-PAA-EPI polymer magnetic nanoparticle targeting chemotherapy on bladder caner was much better than that of conventional chemotherapy.We believe that targeting chemotherapy has a good application future in bladder caner therapy.
