摘 要:目的和方法:为了探讨巨核细胞增殖调控,我们研究了肝素对重组人血小板生成素(recombinanthumanthrombopoietin,rhTpo)刺激BALB/c小鼠巨核系祖细胞(CFU-MK)和人巨核细胞系-红白细胞白血病(humanerythroleukemia,HEL)细胞株增殖的作用。采用半固体和液体培养、CFU-MK-乙酰胆碱脂酶染色和5-溴-2′-脱氧尿嘧啶核苷(5-BrdU)-酶联免疫吸附等方法,观察了CFU-MK和HEL细胞集落数以及HEL细胞的吸光度。结果:在培养基中加入rhTpo100ng/mL和肝素25U/mL,细胞集落和吸光度高于不加肝素对照组。CFU-MK被扩增150%,HEL细胞集落和HEL细胞吸光度增幅分别为102%和106%。结论:肝素能增强血小板生成素对巨核细胞增殖的作用。AIM and METHOD:In order to investigate the regulation of megakaryocyte proliferation, action of heparin on stimulating the proliferation of BALB/c mice CFU-MK and human megakaryocytic lineage-HEL cells by thrombopoietin (Tpo) were studied. Semi-solid and liquid cultures, CFU-MK acetylcholinesterase staining and 5-bromo-2'-deoxy-uridine (5-BrdU)-elisa were used. CFU-MK, HEL colonies and absorbance were observed. RESULTS: In the medium treated with rhTpo 100 μg/L and heparin 2.5×10 3 U/L, the colonies and the absorbance increased as compared with control group without heparin. A combination of rhTpo 100 μg/L with heparin 2.5×10 3 U/L increased CFU-MK by 150%, HEL colonies by 102% and HEL absorbance by 106%, respectively. CONCLUSION: Heparin enhances the effect of Tpo on the proliferation of megakaryocytes.
