几丁糖-顺铂缓释药膜对裸鼠腹腔种植胃癌细胞生长的抑制作用

摘　　要：目的观察几丁糖-顺铂缓释药膜对裸鼠腹腔种植胃癌细胞生长的作用。方法将几丁糖与顺铂混合,制备几丁糖-顺铂缓释药膜;32只裸鼠腹腔接种胃癌细胞后分为4组,分别植入几丁糖-顺铂缓释药膜、几丁糖膜、顺铂和0.9%氯化钠溶液。结果几丁糖-顺铂缓释药膜具有良好的载药量、药物包封率和体外缓释性;与其他3组比较,可明显减少腹膜癌结节数(P<0.01),提高裸鼠的生存率,延长生存时间(P<0.05),且药物的不良反应轻(P>0.05)。结论几丁糖-顺铂缓释药膜能抑制裸鼠腹腔种植的胃癌细胞生长,延长裸鼠生存时间。Objective To investigate the effects of Chitosan/cisplatin drug release films on the cell grouth of peritoneal dissemination gastric cancer in nude mice.Methods The chitosan/cisplatin mixing chitosan and cis-platinum.32 nude mice inoculated intraperitoneally with gastric cancer cells were divided into four groups.The chitosan/cisplatin films,chitosan membrane,cisplatin and saline were implanted in the mice.Results Chitosan/cisplatinum drug release films showed satisfactory drug loading,drug encapsulation efficiency and in vitro sustained release,Compared with the other 3 groups,Chitosan/cisplatinum drug relense film exhibited significant a decrease in the number of peritoneal nodules（P〈0.01）,an increased in the survival rates of the nude mice and a extension of survival time（P〈0.05）,and a decrease in the side effects of the drug（P〉0.05）.Conclusion Chitosan/cisplatin drug release film can inhibit the peritoneal cells of gastric cancer grown in nude mice and can prolong the survival time of the nude mice.
