摘  要:目的研究15-脂氧合酶-1(15-LOX-1)对胃癌细胞AGS细胞周期的影响,并探讨其作用机制。方法实验分为15-LOX-1重组质粒转染组、空载体组和AGS组,转染胃癌细胞AGS,用RT-PCR和Western blot检测15-LOX-1 mRNA和蛋白的表达,流式细胞术检测AGS细胞周期的分布,Western blot检测S相激酶相关蛋白2(S-phase Kinase associated protein 2,Skp2)和细胞周期抑制因子P27的表达。结果重组质粒转染组AGS胃癌细胞分别从mRNA和蛋白水平检测到15-LOX-1的表达,流式细胞仪结果显示15-LOX-1可使肿瘤细胞大部分滞留于G0/G1期,进入S期的细胞减少,S期比例下降,并且转染15-LOX-1后P27表达上调,而Skp2表达下降(P<0.05)。结论 15-LOX-1可能通过调节细胞周期蛋白Skp2和P27的表达,使细胞周期阻滞于G0/G1期,抑制肿瘤细胞增殖。Objective To investigate the effect of 15-Lipoxygenase-1(15-LOX-1) on cell cycle of gastric carcinoma and to evaluate its mechanism.Methods The human gastric carcinoma cell line AGS were divided into three groups: 15-LOX-1 transfected group,empty plasmid transfected control group and liposome treated group.The expression of 15-LOX-1 mRNA and protein were detected by semiquantitive RT-PCR and Western blot,respectively.The distribution of cell cycle was evaluated by flow cytometry(FCM).The expression of S-phase Kinase-associated protein 2(Skp2) and P27 were detected by Western blot.Results Compared with the empty plasmid transfected control group and liposome treated group,RT-PCR and Western blot both showed the expression of 15-LOX-1 was detected from 15-LOX-1 transfected group.The FCM results shows that 15-LOX-1 could resort a majority of gastric carcinoma cell AGS in period G0/G1 rather than enter the period S,so that the proportion of S period fell.In addition,P27 was up-regulated,meanwhile,Skp2 was down-regulated(P0.05).Conclusion 15-LOX-1 can effectively regulate the expression of Skp2 and P27 to resort gastric carcinoma cell in period G0/G1 and inhibit carcinoma cell proliferation.
