摘  要:目的:探讨雌激素对人肝癌细胞株HepG2细胞载脂蛋白(Apo)AⅠmRNA表达的影响。方法:分别将0.01、0.1、1.0、10.0、20.0μmol/L的雌二醇(E2)与体外培养的HepG2细胞共同孵育24h;选用10μmol/L的E2与HepG2细胞共同孵育0、1、6、24、48h;收集细胞提取总RNA,采用逆转录聚合酶链反应(RT-PCR)测定ApoAⅠmRNA表达水平。结果:随E2浓度的增加,HepG2细胞内ApoAⅠmRNA表达呈现先升高后降低的趋势,以10μmol/L时表达最高,与空白组及0.01、0.1、1.0μmol/L组相比差异有统计学意义(P<0.05),20μmol/L时ApoAⅠmRNA表达降低;时间刺激试验显示随刺激时间的延长,HepG2细胞中ApoAⅠmRNA表达呈现先升高后降低的趋势,刺激24h后表达水平最高,48h后下降。结论:E2可在转录水平调节ApoAⅠ基因表达,并呈现部分的剂量和时间依赖性。Objective:To observe the effect of estrogen on modulating apolipoprotein A(ApoA) Ⅰgene transcription in HepG2 cells.Methods:HepG2 cells were exposed to different concentrations of estradiol(0.01,0.1,1.0,10.0 and 20.0 μmol/L) for 24 h,respectively.HepG2 cells were incubated with estradiol at 10 μmol/L concentration for 1,6,24 and 48 h.RNA was extracted from HepG2 cells and collected at above time points.The expression of ApoAⅠmRNA was analysed by RT-PCR.Results:The level of ApoAⅠmRNA expression was increased with estradiol in a concentration-dependent manner in HepG2 cells.The level of ApoAⅠ mRNA was significantly higher in 10 μmol/L of estradiol than that in 0,0.01,0.1 and 1.0 μmol/L of estradiol.The expression of ApoAⅠmRNA was decline at 20 μmol/L.The level of ApoAⅠmRNA showed a time-dependent increase as stimulated by estradiol in time-course experiment.The expression of ApoAⅠmRNA was significantly increased at 6 h(P 0.05) and the highest at 24 h(P 0.05) compared with that of other time points.Conclusion:Estrogen may play an important role in the transcription of ApoAⅠgene.The effect of estrogen on the mRNA expression showed a concentration-dependent and time-dependent manner.
