槐耳清膏抑制肝癌切除术后复发瘤的生长及机制

摘　　要：目的 探讨槐耳清膏对原位移植瘤手术切除后复发肿瘤的作用及其机制.方法 人高转移肝癌HCC-LM3原位移植瘤建立后14 d,手术切除原位瘤,分别使用槐耳清膏和生理盐水灌胃28 d后,比较肝脏复发肿瘤的大小和肺转移率,免疫组织化学检测肿瘤微血管密度,实时荧光定量聚合酶链反应（PCR）检测肿瘤里血管生成因子表达的变化,酶联免疫吸附试验（ELISA）检测血清血管生成因子的表达.结果 槐耳清膏可显著延缓移植瘤手术切除后复发肿瘤生长、抑制肺转移,降低肿瘤MVD,降低血管生成因子的表达.结论 槐耳清膏显著抑制人肝癌HCC-LM3原位移植瘤手术切除后复发肿瘤的生长以及肺转移,抗血管生成作用可能是其主要机制.Objective To investigate the inhibitory effect of huaier on the growth of recurrent tumor in nude mice, transplanted with orthotopic human hepatocelllular carcinoma HCC-LM3, which underwent hepatic resection and explore the underlying mechanism. Methods Fourteen days after transplantation with orthotopic HCC-LM3 with highly metastatic potential which was transfected with red-fluorescent protein （RFP） gene, nude mice were subjected to hepatic resection, divided into two groups randomly and then treated with huaier 4.5 g/（ kg·day） and normal saline respectively for 28 days. Nude mice were sacrificed and the volume of recurrent tumor and lung metastases in two groups were compared. The microvessel density （MVD） of tumor was observed by immunohistochemistry. The expression of vascular endothelial growth factor （VEGF） and platelet derived growth factor （PDGF） in tumor was detected by real-time reverse transcription-polymerase chain reaction （RT-PCR）, and the expression of serum VEGF and PDGF was examined by enzyme-linked immunosorbent assay （ELISA）. Results Huaier significantly inhibited the growth of recurrent tumor and the lung metastases as compared with control[（0.10 ± 0.05 ） cm3 vs （0.89 ±0.46） cm3, 2/12 vs 10/12 ,P 〈0.05]. The average number of metastatic nodules in lung in huaier group and control group was （0.17 ± 0.41 ） and （ 1.67 ± 1.21 ）, respectively （ P 〈 0.05 ）. Huaier decreased the MVD （2.59% vs 4.55%, P 〈 0.05 ） and the expression of angiogenic factors （ VEGF and PDGF） in tumor and serum. Conclusion Huaier inhibited the growth of recurrent tumor and the lung metastases possibly by its antiangiogenic effect.
