摘 要:目的研究偏侧咀嚼后大鼠咬肌Ca2+质量分数、Ca2+-ATP酶活性以及咬肌超微结构的变化。方法 2009年9—10月于山东大学动物实验中心选取8周龄雌性Wistar大鼠10只,随机分为对照组和偏侧咀嚼组,每组各5只。偏侧咀嚼组大鼠拔除左侧上颌磨牙;对照组大鼠模拟拔牙操作,不拔除牙齿。4周后处死各组大鼠,切取浅层咬肌组织。采用火焰原子吸收分光光度法测定咬肌组织中Ca2+质量分数;Ca2+-ATP酶活性运用超微量Ca2+-ATP酶试剂盒测定;透射电镜观察咬肌组织超微结构的变化。结果偏侧咀嚼组大鼠拔牙侧Ca2+质量分数较非拔牙侧及对照组均有明显升高(P<0.05);偏侧咀嚼组大鼠拔牙侧Ca2+-ATP酶活性较非拔牙侧及对照组均有明显降低(P<0.01)。透射电镜下观察发现,偏侧咀嚼组大鼠咬肌组织的超微结构与对照组相差较大,损伤表现拔牙侧重于非拔牙侧。结论偏侧咀嚼后大鼠拔牙侧咬肌Ca2+质量分数升高、Ca2+-ATP酶活性降低以及咬肌超微结构出现病理性变化。Objective To study the influence of unilateral chewing on Ca^2+ concentration and Ca^2+-ATP enzyme activity and ultrastructure in rat masseter muscle. Methods The 10 rats were randomly divided into(A)normal control group and(B)unilateral chewing experimental group. The Ca^2+ concentration was determined by atomic absorption spectrophotometry;the activity of the Ca^2+-ATP enzyme was determined by super-micro volume Ca^2+-ATP enzyme Kit. The ultrastructure of the rat masseter muscle was observed under TEM. Results The Ca^2+ concentration of the extraction side of group B had significant difference compared with the non-extraction side(P = 0.02 0.05),group A(P 〈0.05);Ca^2+-ATP enzyme activity of group B extraction side were higher than its non-extraction side (P 〈0.01)in group A(P 〈0.01). The ultrastructural changes of the rat masseter muscle under TEM were:the extraction side of group B had more severe pathological manifestations than its non-extraction side. Conclusion Higher Ca^2+ concentration,lower Ca^2+-ATP enzyme activity and pathological changes of ultrastructure appear in rat masseter muscle after unilateral chew.
