5-氟脲嘧啶、丝裂霉素和表阿霉素诱导人胃癌细胞凋亡的体外研究

摘　　要：目的了解5-氟脲嘧啶、丝裂霉素和表阿霉素分别及联合对人胃癌MGC－803细胞凋亡的诱导作用。方法用一定剂量的5-氟脲嘧啶。丝裂霉素和表阿霉素分别及三药联合与人胃癌细胞株MGC－803共同孵育12h后，应用光镜进行细胞形态学观察。细胞DNA经特异性荧光染色后，用流式细胞仪分析细胞周期变化。结果人胃癌MGC－803细胞经5-氧脲嘧啶、丝裂霉素和表阿霉素作用后，出现典型的细胞凋亡形态学改变，定量分析凋亡细胞发生率分别为10．2％、9．1％和9．7％，而三药联合作用后凋亡细胞的发生率为21．4％。结论5-氟脲嘧啶、丝裂霉素和表阿霉素可诱导人胃癌细胞林凋亡，三药联用可提高凋亡细胞的发生率。Objictive To investigate the induction of apoptosis by fluril(5-Fu),mitomycin(MMC)and epirubicin (EPE)in gastric carinoma cell line.Methosd Ahuman gastric carcinoma cellline MGC-803 was cultured in fall medium with 5-Fu,MMC and EPIin gastric carcinoma cell line.Methods Ahuman gastric carcinoma cell line MG-803 was cultured in fall medium with 5-Fu,MMC and EPI for 12hours .The treated cells were examined under light microscope.Cell cycle analysis was peformed on flow cytometry(FCM).Results After treatment with 5-Fu,MMC and EPI,the cells became smaller and condensed.Apo-ptotic bodies were observed.There wer apoptotec peaks in cell cycle analysis on FCM.The percentages of apoptotic cell were respectively 10.2%,9.1%and 21.4% after being treated with 5-Fu,MMC,EPI and threemedicines for 12 hours.Conclusion 5-Fu,MMC and EPI may induce apoptosis in the gastric carcinoma cell ling MGC 803
