胃癌组织Survivin表达及其与凋亡的关系

摘　　要：目的:研究胃癌组织中Survivin表达及其与凋亡的关系。方法:应用免疫组织化学S-P法检测20例正常胃粘膜及120例胃癌组织中Survivin的表达;采用脱氧核糖核酸末端转移酶介导的缺口末端标记(TUNEL)技术,检测其凋亡指数。结果:20例正常胃粘膜组织Survivin均不表达,胃癌组织中Survivin阳性率为64.2%(77/120),二者有显著性差异(P<0.01);Survivin表达与组织分化程度和TNM分期密切相关(P<0.05)。胃癌组织凋亡指数(AI)和TNM分期、组织分化程度密切相关(P<0.05)。Survivin表达水平与凋亡指数呈显著负相关(P<0.01)。结论:Survivin在胃癌中过表达,Survivin在胃癌的发生发展过程中可能起重要作用,检测Survivin和凋亡指数有利于判断胃癌的恶性程度,胃癌凋亡的抑制可能与PTEN蛋白表达上调有关。Objective：To study the expression of Survivin in gastric carcinoma and the relationship between Survivin ex- pression and apoptosis in the tumor. Methods：The expression of Survivin was examined with immunohistochemieal S-P method in 20 specimens of normal gastric mucosa and 120 specimen of gastric carcinoma. The apoptosis index（AI） was examined with terminal deoxymueleotidyl transferase mediated dUTP nick end labeling（TUNEL） technique. Results： All normal mueosas were negative for Survivin. In the 120 gastric carcinomas. 64.2% （77/120） were postive for Survivin, Tile expression of Survivin was well associated with histological differentiation and TNM stage （ P 〈 0.05 ）. The apoptosis index（AI） of gastric carcinoma ceil was negatively associated with TNM stage and histological differentiation （ P 〈 0.01）. The expression of Survivin was negatively correlated with the AI of gastric carcinoma cell （P 〈 0.01 ）. Condusion：Survivin was overexpressed in gastric cancer. Survivin may play an important role in the course of tumorigenesis and progres- sion in gastric carcinoma. It may be valuable to evaluate the malignant degree of gastric carcinoma. The inhibition of ap- optosis in gastric carcinoma could be related with the up - expression of Survivin.
