摘  要:目的研究IL-1β、IL-6及TNF-α在慢性前脑缺血致血管性痴呆(VD)发病机制中的作用。方法采用双侧颈总动脉永久结扎方法制备慢性前脑缺血动物模型;采用免疫组织化学方法检测痴呆鼠脑组织中IL-1β、IL-6及TNF-α的经时变化。结果双侧颈总动脉永久结扎后24h颞叶皮质及海马区的神经元、星形胶质细胞和小胶质细胞即出现IL-1β、IL-6及TNF-α的表达,短期内呈现表达高峰,之后有所下降,至术后4个月时表达仍明显高于对照组,呈持续性高表达。结论炎性因子IL-1β、IL-6及TNF-α可能在慢性缺血致血管性痴呆的发病中起了一定的作用。Objective To study the role of IL-1β,IL-6 and TNF-α on the pathogenesis of chronic forebrain ischemia inducing vascular dementia(VD).Methods We adopted the permanent occlusion of bilateral carotid arteries in male Wistar rats to produce the chronic forebrain ischemia to imitate the human chronic cerebral ischemia.The changes of IL-1β,IL-6 and TNF-αalong with time were detected by immunohistochemical method.Results The IL-1β,IL-6 and TNF-α expression increased on neuron,astrocyte and microglia in hippocampus and temporal lobe 24h after operation,then decreased lightly,while had still been higher until 4 months post operation.Conclusion Inflammatory factors such as IL-1β,IL-6 and TNF-α might be one of the mechanism of chronic ischemia inducing vascular dementia.
