摘  要:目的通过检测戊四唑(PTZ)致痫大鼠钾通道Kv1.2蛋白表达,探讨钾通道Kv1.2与癫痫发病的相关性。方法 40只SD大鼠分成实验组30只和正常对照组10只。实验组30只大鼠通过腹腔注射PTZ建立全身强直阵挛发作大鼠癫痫模型,取成功致痫鼠24只均分成3组,分别于致痫后3个时间段(1h、24h、48h)取脑组织。用免疫组化法和Western blot法检测大鼠钾通道Kv1.2蛋白。结果实验组大鼠海马区钾通道Kv1.2蛋白表达水平在致痫后3个时间段(1h、24h、48h)均明显低于对照组(P<0.05)。实验组大鼠海马区钾通道Kv1.2蛋白表达水平在致痫后3个时间段(1h、24h、48h)之间无显著性差异(P>0.05)。结论大鼠海马区钾通道Kv1.2表达的减少与全身强直阵挛发作大鼠癫痫发病密切相关。Objective To explore the relevance of potassium channels Kv1.2 to epilepsy by detecting expressions of potassium channel Kv1.2 in hippocampus of pentylenetetrazol(PTZ)-induced seizure rats.Methods 40 SD rats were divided into 30 rats of experimental group and 10 rats of normal control group.The epilepsy rat model with generalized tonic-clonic seizure was established by injection of PTZ to 30 SD rats of experimental group.24 epilepsy rats were divided into 3 groups in average and brain tissues were got at different time points(1h,24h and 48h).Immunohistochemistry and Western blot were used to detect expressions of potassium channel Kv1.2 in brain tissue of experimental and normal control group.Results There was obvious decrease of potassium channel Kv1.2 in hippocampus in epilepsy rats at different time points(1h,24h and 48h)than those of normal group(P0.05).There was no obvious difference of potassium channel Kv1.2 in hippocampus of epilepsy rats at different time points(1h,24h and 48h)(P0.05).Conclusion The decrease of potassium channel Kv1.2 in hippocampus of rats may be the important factor of generalized tonic-clonic seizures.
