摘 要:目的通过研究阿托伐他汀对血管性痴呆大鼠海马区突触结构的影响,探讨阿托伐他汀可能的保护机制。方法 830只SD大鼠随机分为假手术组(10只)、模型组(10只)、治疗组(10只),采用双侧颈总动脉永久结扎法制作大鼠血管性痴呆模型,治疗组给予阿托伐他汀5mg.kg-1.d-1(1ml)灌胃;假手术组及模型组予以等量生理盐水灌胃4w。采用Morris水迷宫法测试大鼠的主动学习记忆能力,HE染色观察大鼠海马区细胞形态结构,透射电镜观察海马区超微结构的变化。结果与假手术组比较,模型组大鼠的学习记忆成绩明显下降(P<0.05);治疗组与模型组大鼠相比较,逃避潜伏期缩短(P<0.05),游泳时间百分比增高(P<0.05),穿过平台次数增多(P<0.05),表明学习和记忆成绩均显著升高。与模型组比较,治疗组突触数量明显增多(P<0.05);突触截面积减小且变性程度减轻(P<0.01);细胞形态学观察显示,治疗组大鼠海马区神经细胞损伤减轻,神经元、突触,线粒体及细胞器等数量增加。结论阿托伐他汀改善血管性痴呆大鼠学习和记忆能力的作用机制是保护海马CA1区突触功能。Objective To investigate the protective mechanism of atorvastatin on vascular dementia(VD),through examing the effects of the synapse structure in hippocampus's region of VD rats.Methods Thirty SD rats were randomly divided into sham-operation group(n =10),model group(n=10) and atorvastatin group(n=10).The model of VD rat was induced by permanent bilateral carotid arteries occlusion.Atorvastatin solution were injected into rat's stomaches for 4 weeks in atorvastatin group,and the same volume of normal saline was lavaged for 4 weeks in the sham-operation group and the model groupThe Morris water maze was adopted to test the ability of the rat's initiative learning ability and memory.HE staining was adopted to observe the pathological feature of hippocampus.Transmission Electron Microscope(TEM)was used to observe the ultrastructure of hippocampus.Results Learning ability and memory of model gruop were declined significantly compared with the sham-operation group(P0.05).Learning ability and memory of atorvastatin group were increased remarkablely,the average escape latency were significantly shortened than that in the model group(P 0.05).The percentage of swimming time and the frequency of finding the platform were increased obviously too(P0.05).The Atorvastatin group exhibited an obvious increase in synapse quantity(P0.05),a decrease in synapse sectional area and degeneration degree(P0.01).The result of morphocytology showed that the injuring of neurons of hippocampus in the atorvastatin group were decreased sharply than the model gruop,and the quantity of neurons,synaptic junction,chondriosome and organelle in hippocampus area increased.Conclusion Atorvastatin is beneficial for the improvement of the synapse structure in hippocampus's CAI region of VD rats.Atorvastatin can improve the ability of learning and memory of VD rats,by preventing peroxidation and inflammatory reaction,increaseing cerebral blood flow.
