斑蝥酸钠对膀胱癌EJ细胞株毒性作用的研究

摘　　要：目的:探讨斑蝥酸钠对膀胱癌EJ细胞株的毒性作用。方法:采用5,10,20μg/ml斑蝥酸钠作用于EJ细胞株2,4,24,48 h后,观察细胞形态改变,MTT法检测药物对细胞生长的影响,流式细胞仪检测细胞增殖周期的改变以及细胞凋亡情况。结果:5μg/ml斑蝥酸钠作用2,4 h后细胞无明显改变,10,20μg/ml作用2,4 h后细胞出现水肿、空泡、核固缩以及细胞溶解现象;药物作用细胞后其抑制率显著增高(P<0.05);细胞周期中G2/M期延长,细胞凋亡明显增加(P<0.05)。结论:斑蝥酸钠对膀胱癌EJ细胞株有明显的毒性作用,通过诱导细胞凋亡的途径抑制细胞生长。Objective： To study the cytotoxic efficacy of sodium cantharidinate against human bladder cancer EJ-cell.Methods： After the bladder cancer EJ-cell was treated with 5,10,20 μg/ml sodium cantharidinate on 2,4,24,48 h,the cytology variation was observed and growth inhibition was estimated using MTT assay,and cell cycle perturbations or apoptosis was evaluated by flow cytometry.Results： It was no injure at 5 μg/ml sodium cantharidinate after 2-4 hours to the bladder cancer EJ-cell,and there were denaturalization and necrosis or collapse and death after 2-4 hours at 10,20 μg/ml.And cell was inhibited and induced apoptosis,G2/M-phase arrest was infused.Conclusion： Sodium cantharidinate had obvious cytotoxic efficacy and inhibited growth to human bladder cancer EJ-cell.
