摘 要:目的:对牙体脱钙和劈牙取髓法在牙髓血管内皮生长因子(vascular endothelial growth factor,VEGF)免疫组织化学研究中的染色效果进行比较,初步探讨牙体脱钙技术在牙髓免疫组织化学研究中的应用。方法:取因正畸治疗需要而拔除根尖孔完全闭合的健康下颌前磨牙30颗,分别通过牙体脱钙和劈牙取髓制作牙体牙髓联合切片和牙髓切片,进行VEGF免疫组织化学染色,在显微镜下观察两者的染色效果和组织结构特点。结果:牙体脱钙法制得的牙体牙髓联合切片组织染色效果良好,牙髓细胞、成牙本质细胞、牙本质小管结构清晰,VEGF阳性染色空间分布定位明确;劈牙取髓法制得的牙髓切片成牙本质细胞残缺不全,组织厚薄不一,染色对比度差。结论:牙体脱钙技术应用于牙髓免疫组织化学研究的组织染色效果良好、结构清晰,能够保全牙髓组织。Objective: To compare immunohistochemical staining results of vascular endothelial growth factor(VEGF) in human dental pulp by decalcification and splitting teeth and explore the application of decalcification in immunohistochemical study of human dental pulp.Methods: Thirty healthy and mature mandibular premolars were collected for orthodontic reasons and studied with VEGF immunohistochemistry staining respectively by decalcification and splitting teeth,produced two different dental pulp sections and to observe the staining results between the two methods under the microscope.Results: Sections processed with decalcification worked well,in which pulp cells,odonoblasts and dentinal tubule structure were clear and VEGF positive distribution were well defined;While,sections dealt with splitting teeth were with incomplete odonoblasts,poor staining and weak contrast.Conclusion: Decalcification applicated in dental pulp immunohistochemical study can keep structure clear,preserve experiment results objective and is well recommended.
