摘  要:目的:研究雷公藤内酯醇(triptolide,TL)对纤维肉瘤HT-1080细胞DNA甲基转移酶(DNA methyltransferase,DNMT)-1、DNMT-3A和DNMT-3B表达的影响。方法:用终浓度依次为6、12、18 nmol/L的TL处理HT-1080细胞72 h,对照组不加药物处理,反转录(RT)-PCR、Western blotting分别检测TL对DNMT-1、DNMT-3A及DNMT-3B mRNA和蛋白表达的影响。结果:与对照组比较,18 nmol/LTL处理组DNMT-1和DNMT-3AmRNA表达[分别为(1.10±0.21)/(0.69±0.19),(0.86±0.20)/(0.43±0.12)]及蛋白表达[分别为(0.93±0.20)/(0.54±0.10),(0.86±0.14)/(0.51±0.18)]均明显上调(P均<0.05),DNMT-3B mRNA和蛋白表达无明显变化(P均>0.05);6 nmol/L和12 nmol/LTL处理组DNMT-1、DNMT-3A和DNMT-3BmRNA及蛋白表达均无明显变化(P均>0.05)。结论:TL上调HT-1080细胞DNMT-1和DNMT-3A的表达,可能是TL上调基因甲基化水平的机制。Objective: To evaluate the effects of triptolide on the mRNA and protein expression of DNA methyhransferases (DNMT)- 1, -3A and -3B in human fibrosarcoma HT-1080 ceils in vitro. Methods:Reverse transcription polymerase chain reaction (RT-PCR) and Western blotting were applied to measure the mRNA and protein expression of DNMTs in human fibrosarcoma HT-1080 cells after 72 hours of treatment with 6 , 12 or 18 nmol/L triptolide, respectively. Results:The mRNA and protein expression of DNMT-1 and -3A was statistically elevated in HT-1080 ceils after 72 hours of treatment with 18 nmol/L triptolide, compared with controls ( 1.10 ± 0.21 vs 0.69 ± 0.19, 0.93 ± 0.20 vs 0.54 ± 0.10, 0.86 - 0.20 vs 0.43 ± 0.12, 0.86 ± 0.14 vs 0.51 ± 0.18, respectively, all P〈0.05), while no significant difference was noted between 6 or 12 nmol/L triptolide treated HT-1080 cells and controls (all P〉0.05). And the mRNA and protein expression of DNMT-3B was not significantly changed in HT-1080 cells after 72 hours of treatment with the indicated concentrations of triptolide, compared with controls (all P〉0.05). Conclusion: Triptolide upregulates the mRNA and protein expression of DNMT-1 and -3A, in HT-1080 cells in vitro.
