摘 要:目的研究橙皮苷(HDN)对非酒精性脂肪肝大鼠的保护作用及其机制。方法脂肪乳剂灌胃﹑饮用蔗糖溶液制备大鼠非酒精性脂肪肝模型。于9周末处死大鼠,测定血清肿瘤坏死因子-α(TNF-α)、肝组织含量和白介素-6(IL-6)水平,免疫组织化学法检测CYP2E1的表达。结果 HDN(160、320mg/kg)组能显著降低模型大鼠血清ALT、AST、FFA、TNF-α、IL-6和肝匀浆MDA的含量,提高SOD含量,同时HDN(320mg/kg)组可使肝组织CYP2E1表达下降。电镜示HDN(160、320mg/kg)组肝细胞内脂滴变小、减少,与模型组相比,线粒体肿胀明显减轻,内膜上嵴粒增多。结论 HDN能明显减轻非酒精性脂肪肝大鼠肝组织脂肪变和炎症程度,减轻脂质过氧化,并能显著降低血清TNF-α、FFA和IL-6水平,减少肝组织CYP2E1表达,抑制肝细胞凋亡,这可能是其有效防治非酒精性脂肪肝的作用机制。Objective To study the protection effect and its mechanisms of hesperidin( HDN) with non-alcoholic fatty liver. Methods The rats of the model group were fed with high fat emulsion and sucrose. HDN groups and tiopronin group were administrated with related does of medicine by the end of the 3th week and for six weeks. All rats were sacrificed at the 9th week end. The serum ALT,AST,TNF-α,FFA,IL-6 and hepatic SOD,MDA were measured respectively. The expression of CYP 2E1 in liver tissue was detected by immunohistological staining. Hepatic histopathology from all the groups were analyzed by HE staining and electron microscope. Results Compared with the model group,levels of ALT,AST,TNF-α,FFA,IL-6,MDA decreased significantly in administration of HDN( 160 ,320 mg / kg) ,meanwhile the activity of SOD were enhanced,CYP2 E1 expression decreased. HDN could significantly alleviate the symptoms in the non-alcoholic hepatopathy rats. Conclusion HDN can remarkably ameliorate fatty degeneration and inflammation of hepatocytes,and decrease lipid peroxidation and the levels of TNF-α, FFA ,IL-6 in serum and liver tissue CYP 2E1 expression. This may serve as one of the effective mechanisms which HDN prevents non-alcoholic fatty liver efficiently.
