摘 要:目的:探讨新生大鼠缺氧缺血再灌注脑损伤(hy-poxic-ischemic reperfusion brain damage,HIRBD)后不同时间脑组织肾上腺髓质素(Adrenomedullin,AM)mRNA、AM及外周血血浆AM水平变化规律。方法:将42只健康7 d龄SD实验大鼠随机分为正常对照组、假手术组、HIRBD后0 h、6h、12 h、24 h、48 h组,每组6只,按时剖杀各组实验大鼠,断头取血及双侧脑组织,采用半定量RT-PCR法测定脑组织AM mRNA,酶联免疫吸附法测定脑组织和血浆AM水平。结果:①HIRBD各组脑组织AM mRNA、AM和血浆AM水平显著性增高,与对照组比较有统计学意义(P均〈0.05),高峰在6~24 h,24 h后开始下降。②脑组织AM mRNA水平和脑组织AM水平呈正相关(r=0.75,P〈0.05)。③外周血血浆AM水平和脑组织AM水平呈正相关(r=0.78,P〈0.05)。结论:缺氧缺血再灌注可诱导新生大鼠脑组织AM基因表达和转录水平的增高,并具有一定的时间依赖性;新生大鼠HIRBD时脑组织AM基因表达和转录水平的增高可能是外周血血浆AM增高的主要原因。Objective:To investigate the mRNA expression of adrenomedullin(AM) in cerebral tissue at different time and variation of AM and plasma AM in neonatal rat model with hypoxic-ischemic reperfusion brain damage(HIRBD).Methods: Forty-two Sprague-Dawley(SD) rats aged 7d were randomized into groups of normal control,sham operation and HIRBD at 0 h,6 h,12 h,24 h or 48 h,respectively(n=6 for each group),and were decapitated for obtaining the blood and brain tissues.Semi-quantitative RT-PCR method was used to determine the mRNA expression of AM in the cerebral tissue and enzyme-linked immunosorbent assay(ELISA),for measuring the levels of AM in cerebral tissue and AM.Results: ①The expression of AM mRNA and the levels of AM in plasma and cerebral tissue were increased significantly in each group,with peak at between 6 h and 24 h,but fall after 24 h,as compared with the controls and sham operation group(P0.05);②The expression of AM mRNA was positively correlated with level of AM in the cerebral tissue(r=0.75,P0.05);③The levels of cerebral tissue AM were also positively correlated with plasma AM(r=0.78,P0.05).Conclusion: Hypoxic-ischemic reperfusion may result in up-regulated mRNA expression and transcription of AM in cerebral tissue of neonatal rats,with certain time-dependence,and the increased mRNA expression and level of AM in the experimental animals with HIRBD appears associated with elevated plasma AM level.
