摘 要:目的探讨降钙素基因相关肽(CGRP)在三叉神经痛(TN)发病机制中的作用。方法 40只雄性Wister大鼠随机均分为实验组(A组)及对照组(C组)。A组大鼠左侧三叉神经分支眶下神经行慢性缩窄环术。观察术后大鼠对机械刺激的反应阈值,三叉神经节(TG)切片行CGRP免疫组织化学反应和原位杂交反应,光镜观察摄片,并用HPIAS-1000病理图文分析系统软件进行测定。结果术后2~8周,A组大鼠左侧疼痛阈值较对侧及C组左侧显著降低(P<0.01),左侧TG内CGRP及CGRPmRNA的表达较对侧及C组左侧显著升高(P<0.01);术后12周差异无统计学意义。结论 CGRP在TN的发病机制中发挥重要的作用。Objective To explore the effect of calcitonin gene-related peptide (CGRP) in mechanisms of trigeminal neuralgia. Methods Forty Wistar male rats were randomly divided into experimental group(A) and control group(C). An unilateral chronic constriction injury (CCI) was produced on the infraorbital nerve (ION), one of the trigeminal nerves, in group A. The pain-related behaviour was observed. The section of trigeminal ganglia underwent immunohistological and situ hybridization study, Light microscope and microscopic photo analytic software were employed to detect the expressions of CGRP and CGRP mRNA in frozen section of TGs. Results Pain threshold decreased and the expressions of CGRP and CGRP mRNA were enhanced from 2 to 8 weeks after operation in group A compared with group C(P〈0.01) and there was no significant difference on the 12th week between two groups. Conclusion CGRP and CGRP mRNA may be involved in the mechanism of trigeminal neuralgia.
