摘  要:目的分析缺氧诱导因子1α(hypoxia-inducible factor-1α,HIF-1α)在喉鳞癌中的表达及与肿瘤细胞增殖、凋亡的关系,探讨其临床病理学意义。方法应用免疫组织化学染色技术检测HIF-1α和Ki-67在86例喉鳞癌及32例正常喉黏膜上皮组织中的表达;脱氧核糖核苷酸末端转移酶介导的缺口末端标记(terminal deoxynucleotidyltransferase-mediated dUTP-biotin nick end labeling,TUNEL)检测喉鳞癌细胞的凋亡情况。结果 HIF-1α在喉鳞癌组织中阳性表达率高于正常喉黏膜上皮组织(P<0.05),HIF-1α阳性表达率与肿瘤临床分期、淋巴结转移有关(P<0.05);HIF-1α阳性组中细胞凋亡指数低于阴性组(P<0.05);细胞增殖指数在喉鳞癌HIF-1α阳性组及阴性组中差异无统计学意义(P>0.05)。结论 HIF-1α可作为评估正常喉黏膜上皮组织癌变的辅助指标及判断喉鳞癌病情进展的生物学标志;HIF-1α表达水平与喉鳞癌细胞的凋亡抑制有关,与肿瘤细胞的增殖无关。Objective To analyze the relationship between hypoxia-inducible factor-1α(HIF-1α)protein expres- sion and cell proliferation as well as apoptosis in human laryngeal squamous cell carcinoma, and to evaluate their elinicopathological significance. Methods Eighty-six cases of LSCC were analyzed about their clinical and pathological data. Immuohistochemical staining technique was undertaken for the detection of the expression of HIF-1α and Ki-67, and terminal deoxynueleotidyltransferase-mediated dUTP-biotin nick end labeling for apoptosis in 86 LSCCs and 32 normal tis- sues. Results In LSCCs, the positive expression rate of HIF-lct was higher than that in the laryngeal mucosa tissues ( P 〈 0.05). The positive expression rate of HIF-1α was correlated with TNM stage and lymph node metastasis (P 〈0.05). The apoptotic index in HIF-1α positive group was significantly lower than that in HIF-1α negative group (P 〈 0.05 ). However, there was no significant difference between HIF-1α positive group and HIF-1α negative group in the proliferation index. Conclusions HIF-1α might play an important role in the progression of laryngeal carcinoma, and could be used in predicting malignant transformation of normal mucous membranes. HIF-1α was overexpressed in LSCCs, which was significantly associated with apoptosis of tumor cells. (Chin J Ophthalmol and Otorhinolaryngo1,2010,10:288-290)
