摘  要:目的:研究模拟高原条件下大鼠牙周炎动物模型龈沟液中细胞因子水平的变化及其与牙周指数的相关性。方法:将80只SD大鼠随机分为4组(缺氧对照组、缺氧实验组、常氧对照组及常氧实验组),每组20只,成功建立大鼠牙周炎模型(缺氧实验组、常氧实验组)后,在常氧与低氧环境(模拟海拔5000m高原、23h/d)下饲养8周,检测AL、PLI、BI等牙周临床指标,用ELISA法分别检测每组大鼠龈沟液中TNF-α、PGE2、IL-8的含量。结果:缺氧实验组龈沟液中TNF-α、PGE2水平明显高于其余各组(P<0.01),且与牙周指数呈正相关(P<0.01),而IL-8显著低于其他3组(P<0.01),与AL、PLI呈负相关(P<0.01),与BI无相关(P>0.05)。结论:模拟高原条件下,大鼠牙周炎模型龈沟液中TNF-α、PGE2水平较平原条件下升高,而IL-8减少,促使牙周组织炎症加重。Objective:To investigate the concentrations of inflammatory cytokines in gingival crevicular fluid(GCF)in a periodontitis rat model of when exposed to hypoxia at high altitude.Methods:Eighty adult Sprague Dawley(SD)rats were divided randomly into 4 groups:Two periodontitis rat model groups(the hypoxia group and the normoxia group)were raised under hypoxia(simulating the altitude 5 000 meters,23 h per day)and normoxia environment separately for 8 weeks.GCF was collected from different groups.ELISA was used to detect the levels of TNF-α,PGE2 and IL-8 in GCF.Results:The level of TNF-α and PGE2 in the hypoxia group were higher than other groups(P0.01).The level of IL-8 was lower than the other groups(P0.01).Conclusion:Under the simulative environment of highland,the level of TNF-α and PGE2 in GCF in a periodontitis rat model are much higher,while the level of IL-8 is lower than that at condition of normoxia environment.The results suggest these changes may aggravate damage to the paradentium in the process of periodontitis.
