摘  要:目的:研究绿茶浸出物与头孢唑啉合用对耐甲氧西林金黄色葡萄球菌(MRSA)的抗菌机制。方法:高效液相色谱法测定头孢唑啉与绿茶及其提取物合用和头孢唑啉单用处理的MRSA和ATCC25923菌体内头孢唑啉的含量。应用不同浓度的绿茶浸出物或绿茶浸出物-氯化钠溶液检测MRSA在低渗和高渗环境中的耐受能力。结果:经过绿茶浸出物、茶多酚、表儿茶素(EC)、表儿茶素没食子酸酯(ECg)和表没食子儿茶素没食子酸酯(EGCg)与头孢唑啉合用处理的MRSA和ATCC25923组菌体内的头孢唑啉的含量高于单用头孢唑啉组。随着绿茶浸出物和氯化钠浓度的上升,MRSA菌的耐受能力降低。结论:绿茶及其提取物促进头孢唑啉在细菌体内蓄积。经绿茶处理后的MRSA的细胞壁可能受损。Objective: To study the antibacterial mechanism of green tea and its extrects in combination with cephazoline on MRSA.Methods: Cephazoline in MRSA treated by cephazoline alone or cephazoline plus green tea leachate were determined by high performance liquid chromatograph(HPLC).MRSA was incubated in different concentrations of green tea leachate or green tea-NaCl solutions.Results: Cephazoline in MRSA and ATCC25923 were higher after treatment with green tea leachate,tea polyphenol,EC,ECg and EGCg in combination with cephazoline than after treatment with cephazoline alone.With increasing of concentrations of green tea leachate or NaCl,MRSA became lower tolerance to the solutions.Conclusions: Green tea and its extracts can cause cumulation of antibiotics in bacterial body,which is duo to destroy of outside structures of MRSA after treatment with green tea.
