Survivin、Caspase-3表达对鼻咽癌放射敏感性的影响

摘　　要：目的探讨Survivin、Caspase-3表达对鼻咽癌发生、发展及放疗后复发的影响,并研究两者间的相互关系。方法采用免疫组化SP法检测31例放疗后复发鼻咽癌、30例鼻咽非角化型鳞癌、30例鼻咽粘膜慢性炎标本中Survivin及Caspase-3的表达情况。结果 Survivin、Caspase-3表达的阳性率,鼻咽粘膜慢性炎分别为23.33%和96.67%,在鼻咽非角化型鳞癌中分别为73.33%和16.67%,在放疗后复发鼻咽癌分别为83.87%、22.58%,两者在三组中的表达,差异有统计学意义(P<0.001),在鼻咽非角化型鳞癌组与放疗后复发组相比差异无统计学意义(P>0.05)。在放疗后复发鼻咽癌中,Survivin与Caspase-3的表达有显著负相关性(r=-0.398,P<0.05)。结论 Survivin的激活以及Caspase-3的失活参与了鼻咽癌的发生发展过程,Survivin在胞核的阳性表达可能与放射抗性相关。Objective To study the expression of Survivin and Caspase-3 as well as their effect on genesis and recurrence of nasopharyngeal carcinoma（NPC） and the relationship between the two factors.Methods Immunohistochemical（IHC） method（SP） was used to examine the expression of Survivin and Caspase-3 in paraffin-embedded speciments of 30 chronic nasopharyngitis,30 NPC and 31 recurrent NPC after treated with radiotherapy.Results The positive rates of Survivin and Caspase-3 protein in chronic nasopharyngitis were 23.33%（7/30） and 96.67%（29/30）,respectively,were 73.33%（22/30） and 16.67%（5/30） in NPC,respectively,83.87%（26/31） and 22.58%（7/31） in recurrent NPC after radiotherapy,respectively,with significant difference in three groups（P〈0.05）.But there was no significant difference between NPC and recurrent NPC after radiotherapy（P〈0.05）.In group of recurrent NPC after radiotherapy,there was significant negative correlation between the expression of Survivin and Caspase-3（r=-0.398,P〈0.05）.Conclusions Survivin and Caspase-3 were main factors of progression of NPC.The positive expression of Survivin in nucli may be correlated with the radioresistence of NPC.
