重楼醇提物抗鼠H22移植瘤血管生成的体内实验研究

摘　　要：目的:探讨重楼醇提物对KM小鼠H22细胞皮下移植瘤的生长及对血管生成的抑制作用。方法:建立小鼠皮下肝癌H22模型,将小鼠随机分为6组,生理盐水20ml/kg阴性对照组、环磷酰胺20mg/kg阳性对照组、重楼醇提物0.5g/kg、1g/kg、2g/kg、4g/kg组,连续灌胃治疗10天,观察用药后肿瘤生长情况,计算抑瘤率,用免疫组化方法测定肿瘤组织中CD34的表达。结果:2g/kg、4g/kg重楼醇提物组瘤重抑制率分别为44.3%和57.4%,与生理盐水对照组相比具有显著性差异(P值分别为0.012、0.001)。2g/kg、4g/kg重楼醇提物组对KM小鼠H22细胞皮下移植瘤治疗后的微血管密度计数与NS对照组比较,差异具有统计学意义(P值分别为0.015、0.001)。结论:重楼醇提物对KM小鼠H22移植瘤具有一定的生长抑制作用,同时可明显抑制肿瘤组织微血管密度,推测其抗肿瘤作用可能与血管生成抑制作用有关。Objective：To approach the inhibitory effect on angiogenesis and tumor growth of H22 hepatoma cell transplantation tumor mouses with alcohol extract of Paridis in vivo.Methods： Established H22 transplantation tumor mouses models and divided randomly into six groups.Then intragastric administration was performed of NS（negative control group）,Cyclophosphamide（positive control group） and alcohol extract of Paridis（0.5g/kg,1g/kg,2g/kg and 4g/kg） to H22 transplantation tumor mouses for ten days.Then observed and measured the growth of tumor,calculated the tumor inhibitory rates and measured the expression of CD34 by immunohistochemistry.Results： Tumor weight inhibitory rates of the groups for 2g/kg and 4g/kg alcohol extract of Paridis were 44.3% and 57.4% respectively,and there was a significant different when compared with NS control group（P=0.012,0.001 respectively）.The expression of microvessel density（MVD）of the groups for 2g/kg and 4g/kg alcohol extract of Paridis also could be suppressed significantly than negative control group in vivo（P=0.015,0.001 respectively）.Conclusion： Alcohol extract of Paridis could inhibit the tumor growth and microvessel density with H22 hepatoma transplantation tumor mouse models.So we could presume the mechanism of antitumor was inhibited tumor angiogenesis.
