摘　　要：目的:研究不同条件下口腔上皮样癌KB细胞及肿瘤球的生长特点。方法:用荧光染料Hoechst33342染色培养在有血清和无血清培养基中的KB细胞,用荧光显微镜观察SP细胞的形态。MTT法绘制KB细胞生长曲线。显微镜观察肿瘤球的生长。结果:用荧光显微镜在有血清和无血清培养基中均观察到KB细胞系中存在浅染和拒染的SP细胞,部分细胞出现偏染现象。酶标仪检测结果显示在有血清和无血清培养液培养的KB细胞,繁殖能力不同。在无血清培养液中可以培育出肿瘤球。结论:有血清和无血清培养液培养的KB细胞生长存在差异,肿瘤细胞可能存在不对称分裂,无血清培养液可培育出繁殖能力强的肿瘤球。Objective：To study growth characteristic of the cancer line BK and its tumor sphere under different condition.Methods：KB cell cultured in serum-supplemented medium and serum-free medium were viewed by fluorescence microscopy after being stained with Hoechst33342.The growth curve of KB cell was made by MTT.The growth of tumor sphere was observed by microscopy.Results：Under-stained SP cells were seen in serum-supplemented medium and serum-free medium.Some of them were not stained in center.The productivity of KB cell cultured in serum-supplemented medium and serum-free medium was different.The tumor sphere was observed in serum-free medium.Concluison：There is difference in KB cell cultured in serum-supplemented medium and serum-free medium.The cleavage of BK cell may be asymmetric.tumor sphere can be generated in serum-free medium.
