DNA修复基因MGMT、XRCC1在食管癌中表达的研究

摘　　要：目的:研究食管癌组织,Barrett食管组织和正常食管黏膜中DNA修复酶MGMT及XRCC1的表达及其意义。方法:用免疫组化法检测食管癌组织、Barrett食管与正常食管上皮中XRCC1与MGMT的蛋白表达情况并且分析它们与食管癌临床病理参数的关系。结果:食管癌组XRCC1蛋白的阳性表达率为89.2%明显高于Barrett食管组的59.1%与正常食管组织的57.1%(P<0.05)。XRCC1的表达与性别,年龄,食管癌的分期,分化程度,浸润深度,肿瘤部位、分型及有无淋巴结转移均无关。食管癌组MGMT蛋白的阳性表达率为46.2%明显低于Barrett食管组的77.2%与正常食管组织的85.7%(P<0.05)。MGMT的表达在食管癌的分期,分化程度,浸润深度,及有无淋巴结转移的临床指标中具有统计学意义,但与肿瘤部位、分型及年龄、性别无关。二者在食管癌中的表达无明显相关关系。结论:XRCC1与MGMT基因在食管癌发生过程中可能起重要作用,XRCC1和MGMT在食管癌中表达无明显相关性,提示两者修复损伤途径相互独立。Objective：To investigate the expression and clinical significance of MGMT and XRCC1 in esophageal carcinoma,Barrett＇esophagus and normal epithelium.Methods： Expression and their relationship with clinicopathological parameters of MGMT and XRCC1 proteins was detected by immunohistochemical techniques in esophageal carcinoma,Barrett＇esophagus and normal epithelium.Results： The positive expression rate of XRCC1 in esophageal carcinoma was 89.2% significantly higher than those in Barrett＇esophagus 59.1%and in normal epithelium 57.1%（P0.05）,and there was no obvious correlation between the expression of XRCC1 protein and age,sex,tumor location,tumor type,tumor stage,infiltrating depth,differentiation degree,lymph nodes metastasis.The expression rate of MGMT in esophageal carcinoma was46.2% lower than those in Barrett＇esophagus 77.2%and in normal epithelium 85.7%,and there were significant differences about the expression of MGMT in the clinicopathological factors,such as differentiation degree,infiltrating depth,lymph nodes metastasis and tumor stage.The expressions of MGMT had no relationship with sex,age,tumor type and tumor location.The expression of XRCC1 and MGMT in esophageal carcinoma had no relashionship.Conclusion： XRCC1 and MGMT genes may play important roles in carcinogenesis and progression of esophageal carcinoma,the expression of XRCC1 and MGMT were not related to each other,they have unattached function.
