摘 要:目的:观察白藜芦醇对模拟日光照射所致皮肤急性光损伤的保护作用。材料和方法:招募11名健康女性志愿者,在每位志愿者背部选取6个非曝光部位。部位1~4分别涂抹白藜芦醇+抗氧化剂、抗氧化剂、白藜芦醇、基质,半小时后予1.5倍最小红斑量的模拟日光照射,部位5仅予照射而不涂抹试验样品(阳性对照),部位6既不照射亦不涂抹试验样品(阴性对照)。每天照射1次,连续照射4天。照射前后测量皮肤EI值。末次照射后24h对受试部位皮肤进行取材,行HE染色,测量表皮厚度并进行日晒伤细胞计数。结果:在阴性对照部位无红斑和日晒伤细胞,EI值、表皮厚度分别为194.81、69.25;在阳性对照和涂抹基质的照光部位均出现明显的红斑,EI值增高、表皮增厚、日晒伤细胞增多,分别为432.09、123.75、20.38;涂抹白藜芦醇+抗氧化剂部位红斑轻微,EI值升高、表皮增厚、日晒伤细胞增多程度显著低于其它照光部位,分别为335.42;91.25;9.13(P<0.05)。结论:白藜芦醇联合抗氧化剂可有效地保护模拟日光照射引起的急性光损伤。Objective To investigate the protective effects of resveratrol on the back skin irradiated by solar simulator ultraviolet irradiation (ssUVR). Materials and methods Eleven healthy female volunteers were enrolled in this study. Six sites on the non-exposed back skin were marked for the experiment. Sites 1~4 were applied with resveratrol (Res)+ antioxidant (Aox),antioxidant,resveratrol,vehicle,and followed by ssUVR at a dosage of 1.5 minimal erythema dose (MED) 30min later. Site 5 received ssUVR but without any test materials (positive control)and site 6 received neither ssUVR nor test materials (negative control). The subjects exposure to ssUVR once a day for 4 days.EI value were assessed pre-and after-irradiation. The skin biopsies were taken 24 hours after the last ssUVR. The specimens were stained with hematoxylin eosin (HE) staining. epidermal thickness (ET) was measured and the number of sunburn cell (SC) was counted under the microscope. Results On negative control site,no erythema and sunburn cell (SC) was observed and EI value ,epidermal thickness (ET) were 194.81 and 69.25 respectively. On positive control and Vehicle+ UV sites,obvious erythema was developed,EI values,ET SC significantly increased to 432.09,123.75 and 20.38 respectively. Whereas on Res+Aox+UV site,only slightest erythema was seen,EI value,ET and SC reached 335.42,91.25 and 9.13 respectively,with the least changes comparing to other irradiated sites (P 0.05). Conclusions Combination of resveratrol and antioxidant or one of them were essential for protection against ssUVR-induced damage.
