摘  要:目的 观察地塞米松对培养的牛眼小梁细胞水通道蛋白-1(AQP-1)表达的影响.方法 应用含浓度为5,25,50,250μg/L的地塞米松培养液作用体外培养的第3~4代牛眼小梁细胞.7d后,免疫组织化学方法检测牛眼小梁细胞在不同浓度地塞米松作用下AQP-1的蛋白表达水平,与正常体外培养的细胞进行对照并进行定量分析.结果 正常牛眼小梁细胞可见AQP-1蛋白表达,其灰度值为184.83±1.52,在浓度为5,25,50,250μg/L地塞米松作用7d后,AQP-1蛋白表达灰度值分别为185.95±1.49,193.25±1.18,196.88±0.91,204.26±1.64.当地塞米松浓度≥25μg/L时出现抑制作用(P〈0.05).结论 在体外培养条件下,地塞米松可抑制牛眼小梁细胞AQP-1的表达,这可能与糖皮质激素应用后房水流出阻力增加、眼压升高所继发的青光眼有关.Objective To evaluate the effect of dexamethasone on the expression of aquaporin-1 ( AQP-1 ) in cultured bovine trabecular meshwork cells. Methods Bovine trabecular meshwork cells were cultured in vitro and reproduced to the third and the fourth generation ,then treated with dexamethasone at the concentrations of 5,25,50,250μg/L respectively for 7 days. The expression of AQP-1 was detected by immunohistochemistry technique in cultured bovine trabecular meshwork cells and those treated by dexamethasone. Results AQP-1 was found in normal bovine trabecular meshwork cells, and the grayscale value was 184.83 ±1.52, while in trabecular meshwork cells treated with 5,25,50,250μg/L concentrations of dexamethason the grayscale value was 185.95 ±1.49,193.25 ±1.18,196.88 ±0. 91,204. 26 ±1. 64. When the concentration of dexamethasone was higher than 25 μ/L, the expression of AQP-1 was signifieantly inhibited ( P 〈 0. 05 ). Conclusion The expression of AQP-1 in bovine trabecular meshwork cells was significantly inhibited by dexamethasone. AQP-1 may be involved in the regulation in the aqueous outflow in glucocorticoid-induced glaucoma.
