摘 要:选取18只健康杂交雌犬,随机分为A、B、C、D、E、F共6个组,每组3只。A、B、C、D、E组均实施子宫卵巢摘除术制作动物疼痛模型;F组不做手术作为对照组。A、B、C组分别一次性皮下注射高乌甲素0.2、0.4、0.8 mg/kg,D组口服卓比林(20 mg/kg);E组不给与镇痛治疗。各组动物均在术前、术毕、术后2、6、12、24 h等时间点进行疼痛评分并采血测定血浆中前列腺素E2(PGE2)含量。结果显示:E组术后PGE2含量随时间增加而显著升高;高乌甲素能显著降低术后血浆PGE2含量(P<0.05),B组效果最佳;D组与B组无显著差异(P>0.05)。术后各组的疼痛评分,D组与B组差异不显著(P>0.05)。结果表明,高乌甲素的镇痛效果与卓比林相当,且其镇痛机理可能是通过抑制PGE2生成来实现的。Eighteen clinical healthy local female dogs were selected to evaluate the analgesia effect of lappaconitine and to reveal its analgesia mechanisms.The dogs were randomly divided into 6 groups: group A,B and C :subcutaneous injection lappaconitine 0.2,0.4 and 0.8 mg/kg only one time;group D:oral administration with tepoxalin 20 mg/kg one time;group E :non-analgesia;group F:blank.Before the ovariohysterectomy was finished in each group except group F,peripheral blood 5 mL were collected from each dog for determining the plasma concentration of PGE2 at the time of 2,12 and 24 h postoperation,and pain scores were recorded meanwhile.The result shows that the postoperative plasma concentration of PGE2 in group E was increased significantly as time goes on.The lappaconitine decreased plasma concentration of PGE2(P0.05) significantly,it is the most effective in group B;there was no significant difference between group B and group D(P0.05).There was no significant change of pain scores between group D and group B(P0.05).The data indicated lappaconitine is as effective as tepoxalin,the dosage of 4 mg per-time used for subcutaneous injection is most effective.The inhibition of PGE2 is additional analgesia mechanisms of lappaconitine.
