多药耐药蛋白P-gp,Topo-Ⅱ与p53蛋白在胃癌组织中的表达及意义

摘　　要：目的研究胃癌组织中多药耐药蛋白P-gp、Topo-Ⅱ,凋亡调控蛋白p53的表达及与临床病理因素间的关系。方法应用免疫组化方法检测82例胃癌组织及癌旁胃黏膜中P-gp、Topo-Ⅱ与p53蛋白的表达,分析其与临床病理特征及相互间的关系。结果胃癌组织中P-gp、Topo-Ⅱ及p53蛋白阳性表达率分别为70.73%,49.51%、56.10%,均显著高于癌旁黏膜;P-gp表达率与胃癌远处转移及TNM分期呈显著正相关(P<0.05);p53蛋白高表达率与淋巴结转移及远处转移呈显著正相关(P<0.05);Topo-Ⅱ和p53蛋白表达在高、中、低分化腺癌组间的差异有统计学意义(P<0.05)。结论 P-gp、TopoⅡ和p53蛋白过表达,与胃癌的生长、分化及转移的生物学行为存在密切关系,可作为判断胃癌恶性程度的生物学指标。Aim To investigate the relationship between expressions of P-gp,TopoⅡand p53 proteins in gastric carcinoma（GC） with clinical pathological features.Methods Immunohistochemical staining for P-gp,TopoⅡ and p53 proteins was performed in 82 cases of gastric carcinoma and adjacent tissues.Results The positive rates of expression of P-gp,TopoⅡand p53 proteins in the GC were 70.73%,49.51%and 56.1%,which were significantly higher than those in adjacent tissues.The expression of P-gp positively correlated with distant metastasis and clinical TNM stage.The positive rates of high expression of p53 protein were markedly higher in patients with lymph lode metastasis or distant metastasis than those without metastasis.There were ststistical difference of expression of TopoⅡand p53 proteins among well-,moderate-and poor-differentiated.Conclusion P-gp,TopoⅡand p53 proteins were highly expressed in the GC,whichproduced multiple effects and associated with some clinical pathological features.It was positively correlated with growth,proliferation and metastasis of gastric carcinoma.
