摘 要:目的检测乳腺癌组织中Notch1受体的表达水平,为乳腺癌的防治提供新的靶点。方法分别应用荧光定量PCR(FQ-PCR)和免疫组化技术检测60份乳腺癌组织中的Notch1受体蛋白及基因表达情况。实验设25份正常乳腺组织作对照。结果 FQ-PCR检测60份乳腺癌组织Notch1基因表达阳性,阳性率为87.5%,对照组为12.0%,DNA拷贝数分别为(3.66±0.37)×106拷贝/ml和(0.57±0.76)×102拷贝/ml,差异有统计学意义(P<0.05);免疫组织化学染色检测乳腺癌组织中Notch1分子表达阳性率为81.0%,正常乳腺组织阳性率为12.0%,差异有统计学意义(P<0.05)。结论乳腺癌组织高表达Notch1受体,表明Notch1受体与乳腺癌的发生、发展密切相关。Objective To detect the expression of Notch1 receptors in human breast carcinoma tissues and provide a new target for breast cancer therapy.Methods Notch1 receptor protein and gene expression were detected by FQ-PCR and immunohistochemistry in 60 breast cancer tissue specimens and 25 normal breast tissue specimens.Results Sixty specimens of breast cancer tissue with Notch1 gene expression were analyzed with FQ-PCR.Cancer specimens were positive for expression at a rate of 87.5% and control specimens were positive for expression at a rate of 12.0%.The number of DNA copies was 3.66±0.37×106/ml and 0.57±0.76×102 /ml,respectively(P0.05).Immunohistochemistry revealed positive levels of Notch1 expression in breast cancer tissues at a rate of 81.0% while levels were positive in normal breast tissues at a rate of 12.0%(P0.05).Conclusion Notch1 receptors are highly expressed in breast carcinoma.Notch1 receptors appear to be closely related to the onset and development of breast carcinoma.
