摘 要:目的观察高迁移率族蛋白B1(HMGB1)对小鼠肝脏的病理作用。方法 BALB/c小鼠分为正常对照组、重组HMGB1组和重组HMGB1加抗HMGB1抗体组,每组5只,腹腔注射重组HMGB1和抗HMGB1抗体的剂量分别为100μg/只和500μg/只,注射后24 h,观察血清丙氨酸氨基转移酶、天门冬氨酸氨基转移酶活力、肿瘤坏死因子-α水平以及肝组织形态病理学的变化。另外,采用腹腔注射高剂量的重组HMGB1(500μg),观察小鼠存活情况。结果注射重组HMGB1后,血清丙氨酸氨基转移酶、天门冬氨酸氨基转移酶活力和肿瘤坏死因子-α水平均明显升高(P<0.01),肝组织显示凝固性局部坏死、血管充血并伴大量炎性细胞浸润;同时使用抗HMGB1抗体处理后,血清上述指标明显下降(P<0.05),组织坏死和炎性细胞浸润明显减轻。5只小鼠注射高剂量重组HMGB1后24、48 h各死亡1只。结论 HMGB1对小鼠肝脏组织细胞具有损伤作用。Objective To study the pathological effect of high mobility group box 1(HMGB1) on mouse liver.Methods The BALB/c mice were randomly divided into normal control group,recombinant HMGB1 group and recombinant HMGB1 plus anti-HMGB1 antibody group.Each group included 5 mice.The changes were investigated for serum activities of alanine aminotransferase and aspartate aminotransferase,serum level of tumor necrosis factor-alpha,and liver histopathological morphology,24 hours after the mice were intraperitoneally injectedwith the dosages of 100 μg recombinant HMGB1 and 500 μg anti-HMGB1 antibody per mouse.In addition,the lethality of high dosage of recombinant HMGB1(500 μg per mouse) to mice was investigated.Results Serum activities of alanine aminotransferase and aspartate aminotransferase,and serum level of tumor necrosis factor-alpha markedly increased(P0.01),after mice were administered with recombinant HMGB1.In addition,local necrosis,vascular hyperaemia and mass inflammatory cell infiltration.were found in the liver tissue.Anti-HMGB1 antibody conferred significant protection against the liver damages induced by recombinant HMGB1.2 of 5 mice were died at 24 and 48 hours after mice were administered with the high dosage of recombinant HMGB1,respectively.Conclusion HMGB1 induces liver damages in mice.
