摘 要:目的研究瘦素对创伤大鼠抗氧化功能的影响,为阐明瘦素促进创伤愈合作用的机制提供实验依据。方法 30只Wistar雄性大鼠随机分成3组:正常对照组、创伤对照组和瘦素治疗组。3组大鼠均用5%Na2S背部脱毛。瘦素治疗组和创伤对照组大鼠背部制做皮肤缺损创面(2.0 cm×2.5 cm),并分别用瘦素蛋白溶液0.1 ml(约含瘦素2.0μg)和等体积生理盐水涂抹,每天1次,连续7 d后处死大鼠,取血、伤口肉芽组织及正常对照组大鼠相应部位脱毛皮肤检测血清和皮肤组织抗氧化指标。结果瘦素治疗组肉芽组织MDA水平〔(0.142 2±0.065 7)mmol/g〕明显低于创伤对照组〔(0.221 2±0.086 1)mmol/g,(P<0.05)〕,接近正常对照组水平〔(0.185 5±0.033 6)mmol/g〕;而瘦素治疗组肉芽组织SOD活性〔(45.91±7.04)Nu/g〕、蛋白含量〔(114.98±7.01)mg/g〕分别明显高于创伤对照组肉芽组织〔(34.15±8.55)Nu/g、(104.01±14.17)mg/g,(P<0.05,P<0.01)〕,接近和低于正常对照组皮肤水平〔(39.54±7.69)Nu/g、(148.60±7.68)mg/g,(P<0.01)〕。创伤对照组与瘦素治疗组大鼠血清MDA、GSH水平及SOD活力差异无显著性。结论瘦素能明显改善创伤组织局部的氧化应激损伤,增强其抗氧化能力,这可能是瘦素发挥促进创伤愈合作用的机制之一。Objective To study the effect of leptin on antioxidation in wounded rats,so as to provide the experimental basis for elucidating the mechanism for accelerating wound healing by leptin.Methods Thirty male Wistar rats,weighed 180±20 g,were randomly divided into three groups: normal depilation group,wound control group,and leptin treatment group.A full-thickness skin defect measuring 2.0 cm×2.5 cm was made at the back of rats in wound control group and leptin treatment group.0.1 ml leptin solution(about 2.0 μg leptin) was applied to the wound of rats in leptin treatment group daily for 7 days,and that in wound control group with equivalent normal saline.All animals were killed and then blood,granulation tissue samples,and normal corrseponding skin were collected to examine the antioxidation.Results MDA level in granulation tissue in leptin treatment group((0.1422±0.0657)mmol/g) were significantly lower than in wound control group((0.2212±0.0861)mmol/g,(P0.05)),and approached that in normal depilation group.SOD activity and protein content in granulation tissue in leptin treatment group((45.91±7.04)Nu/g,(114.98±7.01)mg/g respectively)were significantly higher than in wound control group((34.15±8.55)Nu/g,(P0.01),(104.01±14.17)mg/g(P0.05)),and approached those in normal depilation group.Serum MDA,GSH level and SOD activity were not different between leptin treatment group and wound control group,suggesting that leptin may have direct action on local wound.Conclusion Leptin can improve the antioxidant ability and attenuate oxidative stress in local wound tissues,which may be part of the mechanism for wound healing effect of leptin.
