摘 要:采用自主研制的微电极芯片系统,研究电脉冲参数及Ca2+对酵母细胞穿孔率的影响.发现酵母细胞穿孔率随着脉冲电压、脉冲持续时间、脉冲个数的增大而升高,且电穿孔具有累积效应,在脉冲电压40V、脉冲持续时间10μs、脉冲个数8个的条件下,穿孔率达到83%.此外,研究了钙离子对酵母细胞穿孔率的影响,一定浓度的Ca2+能提高酵母细胞穿孔率,当Ca2+浓度为0.1mmol/L时,穿孔率可达到89%;Ca2+浓度过高会降低酵母细胞的穿孔率甚至会抑制穿孔的发生,并从机理方面对其进行了初步探讨,为进一步研究电穿孔机制提供有益参考.The impact of electrical pulse parameters and Ca2+ on the electroporation ratio of yeast cells was studied on a homemade microelectrode array based chip system.The electroporation ratio was observed to climb up with the increase of the electric voltage,duration,and number of the pulse.Furthermore,there is a cumulative effect in the electroporation process.When the electric voltage,duration and number of the pulses were 40 V,10 μs,and 8,respectively,the electroporation ratio could reach 83%.The impact of calcium ion on the electroporation ratio was also studied.The results showed that a certain concentration of Ca2+ could raise the electroporation ratio.For example,the electroporation ratio could be up to 89% when the concentration of Ca2+ was 0.1 mmol /L.However,too high concentration of Ca2+ might reduce the electroporation ratio even inhibit the occurrence of the electroporation.Its mechanism has also been preliminary studied and could provide a useful reference for further study.
