摘 要:目的:建立蛹虫草子实体及其培养基中虫草素(Cordycepin)含量的测定方法。方法:采用高效液相色谱(HPLC)法,选用YMC-PackC18色谱柱(250×4.6mm,5μm),流动相为甲醇-水(20:80),流速为1mL/min,检测波长为260nm,柱温为30℃,分析时间为20min。结果:蛹虫草子实体中虫草素含量为0.503mg/g,平均加样回收率(n=9)为100.8%(RSD=1.33%);蛹虫草培养基中虫草素含量为0.118mg/g,平均加样回收率(n=9)为101.7%(RSD=1.82%)。结论:本方法分离度好、精密度高、专属性强、灵敏度高,可用于蛹虫草子实体及其培养基的质量评价。Objective:A new HPLC method was developed to determine the content of the Cordycepin in fruitbodies as well as Culture medium.Methods:HPLC was performed on a YMC-Pack C18 reversed-phase column(250×4.6mm,5μm) .All of the reference substances and sample were separated with the mobile phase of methanol:water(20:80) under isocratic elution for 20min,flow rate was 1.0mL/min,the detection wavelength was 260nm,and the column temperature was 30℃.Results:The content of the Cordycepin in the fruitbody were 0.503mg/g and 0.118mg/g in the Culture medium.The average recovery rate were about 101.1%(n=9,RSD=1.33%) in the fruitbody and 101.7%(n=9,RSD=1.82%) in the Culture medium.Conclusion:This developed method,which is simple and with high sensitivity as well as selectivity,can be used for quality evaluation of the Cordyceps.
