摘 要:目的研究拟胆碱药卡巴胆碱对缺血再灌注大鼠血浆TNF-α、IL-6水平的影响及其意义。方法成年雄性Wister大鼠戊巴妥纳腹腔麻醉,用动脉夹夹闭肠系膜上动脉起始部,60min后,松夹恢复肠系膜血流,制成肠缺血-再灌注损伤模型。动物按随机数字表法分为治疗组和对照组。治疗组在夹闭后30min和再灌注后30min,分别肌肉注射卡巴胆碱(0.1mg/kg),对照组则注射等量生理盐水。分别于夹闭后不同时间点测血浆TNF-α、IL-6水平。结果不同时间点TNF-α、IL-6均有下降,两组相比差异有统计学意义。结论卡巴胆碱能抑制缺血再灌注大鼠体内促炎细胞因子的产生,有助于减轻缺血再灌注后全身性炎症反应。ObjectiveTo study the significance of a cholinergic drug carbachol on the levels of TNF-αand IL-6 in ischemia-reperfusion rat plasma.MethodsAdult male Wister rats were anaesthetized by intraperitoneal injection sodium pentobarbital.The proximal part of superior mesenteric artery was occluded with artery clip.After 60 minutes,the clip was removed to restore mesenteric blood flow,and intestinal ischemia-reperfusion injury model was made.Animals were randomly divided into treatment group and control group.The treatment group was intramuscularly injected with carbachol(0.1 mg/kg) 30 minutes after occlusion and 30 minutes after reperfusion,respectively,while the control group was injected normal saline.Plasma TNF-α and IL-6 were measured at different time points after occlusion.ResultsAt different time points,TNF-α and IL-6 decreased in both groups,the difference was statistically significant between the two groups.ConclusionCarbachol inhibits pro-inflammatory cytokine production induced by ischemia-reperfusion in rats,helps to reduce systemic inflammatory response caused by ischemia-reperfusion.
