高频重复经颅磁刺激对大鼠神经元及血脑屏障超微结构的影响

摘  要:目的:研究高频重复经颅磁刺激(rTMS)对大鼠神经元及血脑屏障超微结构的影响。方法:健康成年SD大鼠14只,随机分为对照组和rTMS组各7只,rTMS组以20 Hz、80%最大输出强度行rTMS,鼠脑中线旁左眼后眦与左耳前缘连线的中点接受最强磁刺激,每天1次,15 min。连续2周后取脑,制作超薄切片,在电子显微镜下观察神经元及血脑屏障的超微结构。结果:在刺激过程中,rTMS组未出现肢体强直、阵挛等异常活动。扫描电子显微镜下发现,rTMS组与对照组比较,神经元,核内染色质聚集边集,粗面内质网附着的核糖体数量和线粒体减少,部分线粒体水肿;毛细血管内皮细胞、基膜及周细胞出现明显水肿,部分神经元及毛细血管内皮细胞失去正常形态和结构。结论:长期高频rTMS可一定程度破坏大鼠神经元细胞器及血脑屏障。提示在将高频rTMS应用于大脑时应谨慎。Objective: To observe the effects of high-frequency repetitive transcranial magnetic stimulation(rTMS) on ultrastructure of neurons and blood-brain barrier in rats.Methods: Fourteen Sprague Dawley rats were randomly divided into two groups: control group and magnetic stimulation group.The animals in magnetic stimulation group were subjected to 20 Hz rTMS for 15 min per day at the intensity of 80% maximum output of stimulator for 14 days.Left motor cortex was selected as stimulation locus.The brains were removed,and the ultrathin sections were made.The changes of neurons and blood-brain barrier ultrastructure were observed under the electron microscopy.Results: During stimulation process,no limb rigidity and clonic seizure occurred.Under the scanning electron microscopy,neurons in rTMS group showed morphological changes: margination of chromatin,reduced number of mitochondria,mitochondria swelling.Blood brain barrier in rTMS group showed: swelling of endothelial cells,basement membrane and pericytes.Some neurons and blood brain barrier lost normal structure and organization.Conclusion: Long-term high-frequency transcranial magnetic stimulation may to some extent damage organelles in neurons and reduce the blood-brain barrier permeability.
