摘 要:目的:观察脉冲强磁场联合姜黄素引起膀胱肿瘤细胞BIU-87凋亡的作用。方法:将体外培养的膀胱肿瘤细胞癌株BIU-87随机分为A、B和C 3组。A组为空白对照组,B、C组均经姜黄素处理,C组处理后置于脉冲强磁场中干预(8T,15HZ),每天2 h。分别于干预后的第1、2、3天时采用显微镜观察肿瘤细胞形态、MTT法检测肿瘤细胞生长抑制率。结果:与A组比较,肿瘤细胞显微镜下处理后的B、C组,均出现典型的凋亡细胞形态,以C组表现更加明显;MTT检测,B、C组肿瘤细胞生长抑制率明显上升(均P〈0.01),且呈时间依赖关系,第1-3天肿瘤细胞增殖抑制率B组明显低于C组(18.6%、40.6%、51.4%与26.9%、50%、61.4%,P〈0.01);结论:脉冲强磁场联合姜黄素可促进膀胱肿瘤细胞的凋亡和抑制其生长。Objective: To approach the apoptosis induced by pulsed strong magnetic field combined with curcumin in human bladder cancer BIU-87 cells.Methods: The human bladder cancer BIU-87 cells were put into 96-hole plates,and randomly divided into curcumin group,pulsed strong magnetic field combined with curcumin group and control group.In pulsed strong magnetic field combined with curcumin group,the cells were exposed to pulsed strong magnetic field for stimulation for 2 h every day.Under the inverted phase-contrast microscopy,the cultured cells were observed.MTT assay was applied to reveal viable cells with absorbance(A) value on the first,second and third day.Results: After experimental treatment,typical apoptotic characteristics of the tumor cells were found in two experimental groups,more remarkable in pulsed strong magnetic field combined with curcumin group.The proliferation inhibition rate of tumor cells in curcumin group at the first,second and third day was 18.6%,40.6%,51.4%,and that in pulsed strong magnetic field combined with curcumin group was 26.9%,50%,and 61.4% respectively.As compared with control group by MTT assay,the A values in two experimental groups were significantly reduced(P〈0.001).As compared with curcumin group by MTT assay,the A values in pulsed strong magnetic field combined with curcumin group were significantly decreased(P〈0.01).Conclusion: Pulsed strong magnetic field combined with curcumin could promote apoptosis and suppress the growth of human bladder cancer BIU-87 cells.
