金雀异黄素对增生性玻璃体视网膜病变中血小板源性生长因子和增生细胞核抗原表达的抑制

摘　　要：目的探讨金雀异黄素对兔外伤性增生性玻璃体视网膜病变(PVR)视网膜组织血小板源性生长因子(PDGF)mRNA和增生细胞核抗原(PCNA)表达的影响。方法建立外伤性PVR模型,分为两组:第1组阴性对照组,第2组金雀异黄素治疗组。每组于制模后第7、14、21、28天分别将2只模型眼球取出制作标本,采用原位杂交法检测视网膜组织PDGF mRNA表达,采用免疫组织化学法检测PCNA表达。结果①外伤性PVR视网膜组织PDGF mRNA和PCNA呈先升高后降低的表达;②金雀异黄素治疗组在第7、14、21、28天时视网膜组织PDGF mRNA和PCNA表达均明显低于阴性对照组,比较有显著差异。结论金雀异黄素能降低外伤性PVR视网膜组织PDGF mRNA和PCNA表达,具有抑制外伤性PVR的潜能。【Objective】 To study the effects of genistein on expression of PDGF and PCNA in traumatic prolifera-tive vitreoretinopathy model.【Methods】Traumatic PVR models were induced in rabbits.The models were divided in 2 groups： 1 group（negative control）;2 group（genistein treatment）.2 eyes were enucleated respectively on the 7 th,14 th,21 st,28 th days after surgery in each group.Expression of PDGF mRNA and PCNA in retina was detect-ed respectively by in-situ hybridization examination and immunohistochemical analysis.【Results】In the negative control Group,the expression of PDGF mRNA and PCNA protein were upregulated at first and then down-regulated.In the Genistein treatment Group,the expression of PDGF mRNA and PCNA protein were obvieously down-regulat-ed than those in control Group at 7 d,14 d,21 d and 28 d,and the difference was significant statistically.【Conclusion】Genistein can obvieously inhibit the expression of PDGF mRNA and PCNA protein in traumatic PVR.Genistein might inhibit traumatic PVR.
