摘  要:为研究酒精对神经胶质细胞损害的作用机制,应用荧光偏振技术探讨了酒精及其代谢产物乙醛对神经胶质细胞中星形胶质细胞和少突胶质细胞膜脂质荧光偏振度(Pr)和脂质流动度(LFU)的影响。结果表明2mmol/L酒精、乙醛并不影响星形胶质细胞、少突胶质细胞膜脂荧光偏振度和流动度,5mmol/L,20mmol/L酒精和乙醛均可影响两种细胞的Pr值,导致荧光偏振度降低和细胞膜脂质流动度增高,且均与酒精和乙醛剂量相关。20mmol/L酒精对星形胶质细胞作用强于少突胶质细胞,20mmol/L乙醛对两种细胞膜脂质荧光偏振度和流动度的影响无显著性差异。酒精和乙醛导致两种神经胶质细胞膜流动性增加,致使两种细胞膜的结构发生了改变,这很可能是酒精所致神经系统发育异常的机制之一。In order to research the mechanism of damage on neurogliocytes induced by alcohol and its metabolites acetaldehyde,in this research,fluorescence polarization technique was used to explore the effects of alcohol and acetaldehyde on astrocytes and oligodendrocytes membrane lipid fluidity in rats.The results showed that 2 mmol/L alcohol and acetaldehyde had no effects on fluorescence polarization and lipid fluidity for two kinds of gliacyte,5 and 20 mmol/L alcohol and acetaldehyde could resulted in polarization(Pr) value decreased and lipid fluidity unit(LFU) increased.There were significant relationships between the Pr values and the levels of alcohol and acetaldehyde.The effects of 20 mmol/L alcohol on astrogliacytes was stronger than that of oligodendrocytes,however,the effects of 20 mmol/L acetaldehyde on two kinds of cells,presented no significant differences in Pr value and the levels of LFU.The alcoholacetaldehydeinduced increases of fluidity resulted in the variances of membrane construction in two kinds of neurogliocytes which might be one of the mechnisms for alcoholinduced developmental abnormatities in nervous system.
